Effects Of Yishenhuatanhuoxue Decoction Combined With Diane-35 On Serum Testosterone(T), Serum Luteinizing Hormone(LH) And Its Receptor’s Expression In Polycystic Ovarian Syndrome Rats

Objective:This experiment is set up to observe the serum testosterone(T), serum luteinizing hormone(LH), and the expression of ovarian granulosa cells AR and LHR of PCOS rat model under the application of Yishenhuatanhuoxue decoction unite Diane 35. To dicuss the mechanism of action of Yishenhuatanhuoxue decoction to the follicular of PCOS rat model,which is investigated for providing theoretical and experimental basis for clinical application.Method:Clean-class breed 66 6-week-old female Wistar rats. After 7 days of adaptive feeding, apply vaginal smears to determine their estrous cycles. The ones with regular screening rat estrous cycles will be included in the experiment. Divide the enrolled 66 rats randomly into groups as follows:blank control group,15;model group,15;chinese medicine group,12;western medicine group,12; chinese medicine combined with western medicine group,12;Give the blank control group 10 ml/kg normal saline by its body weight,while give the surplus four groups with Letrozole-carboxymethyl cellulose solution which has concentration of 0.125 mg/m L by gavage, according to 1 mg/(kg/d) per dose in regular daily time continuing for 21 days to create Letrozole induced PCOS rate model.After 21 days,blank control group and model group randomly executed five rats respectively, confirm the model successfully by observe the serum testosterone(T), serum luteinizing hormone(LH) and the morphology of ovary under the microscope. After successfully created,each group was given normal saline, Yishenhuatanhuoxue decoction, Diane 35, and Yishenhuatanhuoxue decoction combined with Diane 35 respectively.Each group is given medications for 15 days which is about 3 estrous cycles. After stopping the medication, monitor an estrous cycle. Fast for 24 hours after the last vaginal smears. Getting blood sample from abdominal aorta, centrifugate to get the serum to set free in the refrigerator for 80 degree Celsius. Dissect the ovariectomized rates and get the wet weight. Rear 4% paraformaldehyde and wait for testing.Result:1. The weight, the bilateral ovarian wet weight and the ovary coefficient of the rat: The weight and the ovary coefficient of the rat in each group hasn’t statistical meaning,while the bilateral ovarian wet weight of the rat has statistical meaning, the wet weights of the ovaries of model group were raised comparing with the blank control group,P < 0.05,which has statistical meaning. Chinese medicine group and chinese medicine combined with western medicine group were dropped comparing with the blank control group, which proves that there is a great difference( P < 0.05),but chinese medicine combined with western medicine group and blank control group had no distinct difference(P > 0.05).2. Ovulation rate: After medical intervention,the ovulation rate of each group has distinct difference(P <0.05), chinese medicine combined with western medicine group had the greatest ovulation rate, following by western medicine group, then chinese medicine group.3. the levels of T and LH: The levels of serum testosterone(T) and serum luteinizing hormone(LH) of model group were raised comparing with the blank control group, P < 0.05, which proves that there is a great difference. The levels of serum testosterone(T) and serum luteinizing hormone(LH) of each group were dropped comparing with the blank control group after medical intervention, P < 0.05, which has statistical meaning. Among them, western medicine group and chinese medicine combined with western medicine group had no distinct difference comparing with the blank control group(P > 0.05),but chinese medicine combined with western medicine group is closer to blank control group.4. The morphology change of ovary: The ovary of model group rats had polycystic-like changes.After medication, the morphologies of the ovaries of each group have improved, Among them, chinese medicine combined with western medicine group is closer to blank control group.5. The expression of ovary AR and LHR: The expression of ovary AR of model group can increase and the expression of ovary LHR can decrease comparing with the blank control group, which proves that there is a great difference( P < 0.05). After medication, chinese m group, western medicine group and chinese medicine combined with model medicine group had distinct difference comparing with the blank control group, P < 0.05, Among them, chinese medicine combined with western medicine group had no distinct difference comparing with the blank control group(P > 0.05), which proves that Yishenhuatanhuoxue decoction combined with Diane 35 is more effective.Conclusion:1.Yishenhuatanhuoxue decoction combined with Diane 35 can improve the ovulation of PCOS rat and the morphology of ovary, decrease the level of ovarian wet weight,serum testosterone(T) and serum luteinizing hormone(LH).2.Yishenhuatanhuoxue decoction combined with Diane 35 can decrease the expression intensity of ovary AR of PCOS rat, and increase the expression of ovary LHR, which is beneficial to the development of follicles and ovulation.3.Yishenhuatanhuoxue decoction combined with Diane 35 is a more effective therapeutic schedule comparing with chinese medicine or western medicine teartment simply.
