Effects Of Zishenyangxinbuxue Decoction On Serum Sex Hormone And The Ovulation Of Diminished Ovarian Reserve Rats

Objective:This experiment is set up to observe the serum estradiol?E₂?,luteinizing hormone?LH?,serum fouicle stimulating hormone?FSH?and the ovulation of DOR rat model under the application of Zishenyangxinbuxue decoction.To initially discuss the mechanism of action of Zishenyangxinbuxue decoction to the DOR rat model,which is investigated for providing the oretical and experimental basis for clinical application.Method:Clean-class breed sixty 12-week-old female Wistar rats.Sixty rats with regular screening rate strous cycles are included in the experiment.After weighing divide the sixty rats randomly into groups as follows:blank control group:15,model group:45.Give the blank control group 10ml/kg normal saline by its body weight,while give the model group with Glycosides of Tripterygium Wilfordii solution which has concentration of 5mg/mL by gavage,according to 50mg/?kg/d?per dose in regular dailytime continuing for 14 days to create DOR rate model.After successfully creating the DOR model,divide the model group into three groups:model control group,western medicine group and chinese medicine group.Blank control group and model control group was given normal saline,chinese medicine group was given zishenyangxinbuxue decoction,western medicine group was given estradiol valerate solution.Each group is given medications for 12 days which is about 3 estrous cycles.After stopping the medication,intraperitoneal anesthesia and get blood sample from abdominal aorta to detect the serum hormone.take the rats?both ovaries for tissue sections and counting antral follicles and luteal.Result:1.The levels of E₂?LH?FSH:The levels of serum fouicle stimulating hormone?FSH?and serum luteinizing hormone?LH?of model group were raised and the level of serum estradiol?E₂?was decline comparing with the blank control group,P < 0.05,which proves that there is a great difference.The levels of serum fouicle stimulating hormone?FSH?and serum luteinizinghormone?LH?of each group were dropped comparing with the model group after medical intervention,P < 0.05,which has statistical meaning.However the level of serum estradiol?E₂?was raised comparing with the model group after medical intervention,P < 0.05,which has statistical meaning.Among them,western medicine group and chinese medicine group had no distinct difference comparing with the blank control group,P > 0.05,and the level of serum estradiol?E₂?between western medicine group and blank control group had no difference,P > 0.05.2.The number of antral follicles and corpus luteum in ovarian tissue luteum:The number of antral follicles and corpus luteum of model group are less than blank control group,P < 0.05,which proves that there is a great difference;chinese medicine group and western medicine group are more than model control group,which proves that there is a great difference;however they are less than blank control group,P < 0.05,chinese medicine group combined with western medicine group is closer to blank control group.Conclusion:1.Zishenyangxinbuxue decoction can effectively decline the levels of LH?FSH of DOR rat,and increase the the level of E₂;2.Zishenyangxinbuxue decoction can improve the ovulation of DOR rat and raise the numberof antral follicles and corpus luteum in ovarian tissue luteum,thereby improve ovarian reserve.
