Background and Objective:Pancreatic cancer is one of the most common alimentary canal neoplasm in China.The mechanism of pancreatic cancer is very complicated and there is no specific clinical symptom and method for diagnosis.Pancreatic adenocarcinoma is one of the most aggressive carcinoma and is very easy to occur vascular invasion,perineural invasion and distant metastases.Because there are no valid indicators of prognosis and the prognosis is very bad.Glioma-associated oncogene 1 is one of the transcription regulators in the Hedgehog signal pathway and is the sign of the activation of the signal pathway.Hedgehog signal pathway plays a important way in the tumorigenesis and the development of neoplasm.Marix metalloproteinase 9 which is also called gelatinase is one member of the Matrix metalloproteinases.Marix metalloproteinase 9 plays a important role in the development of pancreatic cancer via distracting gelatins in the basement membrane and extracelluar stroma and introducingneovascularization.The objective of this study is to observe the expression and its clinical pathological significances of glioma-associated oncogene homolog(GLI-1) and matrix metalloproteinases-9 in pancreatic carcinomas.Material and methods:We collected 42 cases of paraffin-embedded pancreatic adenocarcinoma tissue and 11 cases of paraffin-embedded normal pancreatic tissues from December 2009 to November 2012 in the People’s hospital of Liaoning provenience and the first hospital of China Medical University.The expression of GLI1 and MMP9 were examined by immunohistochemical technique.The relationship between the expression and clinical pathological behaviors were also analyzed.Result:(1)The expression numebers of GLI1 were 32 in pancreatic carcinomas tissues and 1 in normal pancreatic tissues,the differences between them had markedly statistical significance(P<0.05),the staining intensity of GLI1 in pancreatic carcinomas was significantly associated with the Tumor size,differentiation,T stage,TNM stage,Perineural invasion,Vascular permeation, the level of pre-therapeutic CA19-9,Lymph node metastasis,but no distinct difference was found in other parameters.(2)The expression numebers of MMP9 were 30 in pancreatic carcinomas tissues and 1 in normal pancreatic tissues,the differences between them had markedly statistical significance(P<0.05),the staining intensity of MMP9 in pancreatic carcinomas was significantly associated with the Tumorsize,differentiation,T stage,TNM stage,Perineural invasion,Vascular permeation,the level of pre-therapeutic CA19-9,Lymph node metastasis,but no distinct difference was found in other parameters.(3)The patients with positive expression of GLI1 and MMP9 had a bad survival.(4)The expression of GLI1 and mmp9 was positively associated in pancreatic carcinomas.Conclusion:GLI1 plays a important role in the development and metastasis of pancreatic carcinoma through mediating the expression of MMP9. To detect the level of GLI1 and MMP9 maybe a useful way to evaluate the prognosis of the pancreatic cancer patients.
