Expression And Clinical Significance Of OPN, CD44v6 And MMP9 In Pancreatic Carcinoma Using Tissue Microarray

Objective:To investigate the expression of OPN,CD44v6 and MMP9 proteins in pancreatic ductal adenocacinoma(PDAC) by high-throughput tissue microarray technique, as well as its relationship with the clinicopathologic features. In addition, the prognostic significance of OPN,CD44v6 and MMP9 expression in PDAC.Methods:The pancreatic adenocarcinoma tissue microarray was composed of 88 specimens of the pancreatic adenocarcinoma and 11 normal pancreatic tissues as control subject. Immunohistochemistry was performed to examine expressions of OPN,CD44v6 and MMP9, the relationship of these subjects was analyzed with spearmen rank correlation. In addition,53 of the 88 cases were follow-up from 2 months to 6 years, in which the survival was analyzed.Results:Positive expression of OPN,CD44v6 and MMP9 in tumors was showed 54(61.36%),56(63.64%),62(70.45%)respectively. The expression of CD44v6 was closely correlated with pathologic grade, TNM stage and lymph node metastasis rather than location of adenocarcinoma and size(P<0.05). The expression of OPN and MMP9 was significantly associated with TNM stage and lymph node metastasis(P<0.05), but no distinct difference was found in adenocarcinoma location, size and pathologic grade(P>0.05). Spearmen rank correlation showed a significant positive correlation was observed among this three indexes(P＜0.05). Univariate analysis revealed that OPN,CD44v6 and MMP9 are prognosticators for PDAC. Multivariate analysis showed that only OPN has independent prognostic significance.Conclusion:Tissue microarray technique is an effective method to detect multiple gene protein expression. The pathogenesis, development and metastasis of pancreatic cancer is associated with OPN,CD44v6 and MMP9. Evently this three proteins play a joint action in development and metastasis of PDAC. The expression of this three ptoteins is a significant prognostic indicator for PDAC.
