Objective:To investigate the association between the expressions of CD44、 CD133、HIF-1α、VEGF and clinical pathological factors in gastric carcinoma, and to explore its significance in judging the prognosis of patients.To analyze the correlation between the expression of stem cell markers CD44, CD133 and HIF-la, VEGF. To investigate the relationship between tumor stem cells and hypoxia.Methods:Westernblot was used to detect the expressions of CD44 and HIF-la in fresh specimens of gastric carcinoma and tumor adjacent normal tissues.Immuohistochemistry was used to detect the expressions of CD44、CD133、 Hif-1α、VEGF in 22 cases of normal gastric mucosa,41 cases of precancerous lesions of gastric cancer tissues and 103 cases of gastric carcinoma.To analysis the correlation between the expression of CD44、CD133、Hif-1α、VEGF with clinical pathological parameters and postoperative survival time.Results:The positive expression rates of CD44, CD133,HIF-la、VEGF in GC was significantly higher than that in normal tissue and intraepithelial neoplasia (P <0.05); The expression of CD44 was significantly statistically associated with the invasive depth, differentiation degree, lymph node metastasis, TNM stage and postoperative survival time (P< 0.05); The expression of CD133 was significantly statistically associated with the invasive depth, differentiation degree, lymph node metastasis, TNM stage and postoperative survival time(P< 0.05).The expression of Hif-1a was significantly statistically associated with invasive depth、differentiation degree, lymph node metastasis, TNM stage and postoperative survival time(P< 0.05). The expression of VEGF was significantly statistically associated with tumor invasive 、lymph node metastasis and postoperative survival time (P< 0.05). the expression of Hif-1 correlated with VEGF expression (r=0.415, p=0.000);The expression of CD44highCD133highwas significantly associated with Hif-1a (χ2=7.817,p=0.005).Conclusions:The expressions of CD44 and CD133 were correlation with the occurrence and development of gastric cancer, which can be used as an important index of invasion and metastasis, prognosis in GC. CD44highCD133high may be used as the stem cell markers of GC. The expression of Hif-1α can be predicted gastric carcinogenesis, development and prognosis,the overexpression of Hif-1α induces tumor angiongenesis and invasion by activating the transcription of the VEGF gene. The characteristics maintaining of gastric cancer stem cell was correlation with hypoxia, but may not be through the Hif-1α-VEGF pathway.
