Objective:To explore the strong relative stability, good consistency, rich new blood vessels and thick scar tissue in animal models of corneal alkali burn.Method:45 adult healthy Japanese white rabbits(45 eyes)were randomly divided into three groups. Corneal alkali injury was induced by lmol/L Na OH soaked filter paper of 10mm outer diameter、4mm inside diameter for 20 seconds (group A, n= 15),and filter paper of 10mm outer diameter、4mm inside diameter for 40 seconds (group B, n=15),and filter paper of 10mm diameter for 40 seconds (group C,n= 15) respectively.Conjunctiva congestion edema,the situation of inflammation, corneal transparency,corneal ulceration, corneal neovascularization, and Corneal neovascularization (CNV) length and area were observed and recorded under slit-lamp biomicroscope on the first month post-operation the second month post-operation、the third month post-operation.Results:1. Morphological observation:Mixed conjunctiva hyperemia and edema was serious in the first month, group A has a lighter degree than the group B, but group B and group C had no significant different; Conjunctiva hyperemia was mild in second month; The conjunctiva form had a stable state in third the month, The conjunctiva of group B and group C leaved varying degrees deformity. Conjunctiva got a stable state and inflammation subsided. The state of conjunctiva has been in the repair period.2.Corneal turbidity as a quantitative index:Three groups have varying degrees of corneal opacity, The second month opacity degrees is generally lower than the first month, the third month corneal opacity degrees tend to be stable. Under the condition of different burning way and burning time, the degree of corneal opacity was tipping: group C is the best, group B is the second and group C was the third.3.The first month after corneal alkali burn, A, B, C three groups of incidence of corneal ulcer and corneal perforation rate and the rate of corneal neovascularization induced were that the group B was higher than the group A (P<0.05), there was no statistically significant difference between the group B and group C (P>0.05).he second month after corneal alkali burn,the group B was higher than the group A (P< 0.05)about incidence of corneal ulcer and corneal perforation rate.Corneal neovascularization induced rate of group B and group C was higher than group A (P< 0.05), and there was no statistical difference between group B and group C (P> 0.05);The second month after corneal alkali burn,Corneal neovascularization induced rate of group C was higher than group B (P<0.05).4.Corneal neovascularization length, area:after the success of the modeling, the first month, blood vessels was in growth of limbus of cornea corneal central, blood vessel length size, capillary was rich, the color red, blood vessels texture was not clear;The second month, the capillary was rich, the color was light than the first month, vascular texture was clear;The third month,part of capillaries was becoming clear texture、closed to the normal capillary and had a stable growth.In A, B, C three groups,there was a statistical analysis of corneal neovascularization in different times.We were found that corneal neovascularization length, area of group B was higher than group A and group C was higher than group B(P<0.05).So we got that new blood vessels length, area was higher than in group A and group C was higher than group B.Conclusion:1.We can successfully established animal model of implantation of artificial cornea alkali burn、the unified model of the degree of burns and standardized experimental factors by lmol/L Na OH soaked filter paper of 10mm outer diameter、4mm inside diameter for 40 seconds (group B),and filter paper of 10mm diameter for 40 seconds (group C).2.Observe A, B, C three groups for three months, compared alkali burns corneal scarring and thick degree, rate of ulcer, perforation rate, CNV length and area in three groups were found that group B and C were more than 50% about confirmed qualified rate, the two methods are suitable for building an implantable artificial corneal scarring of corneal alkali burn model.3.In the third month, conjunctival congestion, the repair of inflammation, corneal scarring, CNV length and area were stable in three groups, the period of corneal alkali burn model is the best time of implantation of artificial cornea.
