Objective To select effective methods for eye alkaline burn by evaluating the effects of supernatanta of homogenized fresh amnion to rabbit corneal epithelial cell and alkaline burn of rabbi cornea. Methods 1. Experiment in vitro:The rabbit corneal epithelial cell were cultured.We observed the effects of supernatanta of homogenized fresh amnion at different concentration (40?80?160μg/ml)and at different hours (24?48 ?96h)on rabbit corneal epithelial cell by tetrazotium(MTT)colorimetric assay. 2. Experiment in vivo:fifteen rabbits (thirty eyes)were randomly divided into control groups(ten eyes )?experiment groups(supernatanta of homogenized fresh amnion ,ten eyes )and bFGF groups(ten eyes ).The model of alkaline burn of rabbit cornea were set up.Corneal angiogenesis and in epithelium were observed with a slit lamp microscope.The ratio of neovascularization area and epithelium defect to cornea's were calculated.The specimen was taken to stain with HE?VG and AgNORs in different time after cautery .The number of the fibroblast was calculated with microscope.The expression of FN in cornea was detected with immunohistochemical assay. Results: 1. Proliferation rate of homogenate amnion on rabbit corneal epithelial cell:Rabbit corneal epithelial cell can proliferate at different hours(24?48?96hour) and at different concentration of supernatanta of homogenized amnion(40μg/ml?80μg/ml?160μg/ml). 2. The healing of epithelium:The healing area of epithelium of experiment groups and bFGF groups was more than control groups(P<0 .05>at first day?second day and third day.The difference was significant at fouth day to seventh day(P<0.01). 3. The corneal neovascularization: The difference of the area ot corneal neovascularization all of three groups was not significance at seventh day and forth day after alkaline burn of rabbit cornea(P>0.5)? 4. At 7 day,the number of fibroblast cells of homogenate amnion was more than the control group (P<0.05)and the number of bFGF group was far more than the control group(P<0.01).At 14 day,the number of fibroblast cells of all three groups had no obvious difference(P>0.05). Conclusion: 1. Supernatanta of homogenized amnion enhanced the proliferation of rabbit corneal epithelial cell effectively with a dose-dependent manner. 2. Supernatanta of homogenized fresh amnion can accelerate the healing of rabbit corneal epithelial 3. Supernatanta of homogenized fresh amnion can restore the corneal stroma 4. Supernatanta of homogenized fresh amnion can't promote and restrain theGrowth of the corneal neovascularization...
