Objective:Quantified by Real Time Polymerase Chain Reaction, research in breast tumor tissue and adjacent normal tissue expression MALAT1, through the analysis of long-chain MALAT1 noncoding RNA genes in breast cancer and the expression level of paracancerous normal tissue,application of methylation specific PCR technology, analysis MALAT1 gene sequences of DNA methylation,discuss the relationship between methylation of MALAT1 and its expression,aimed to investigate the relationship between MALAT1 gene expression and its methylation status and its potential role in clinical signaling in breast cancer and adjacent noncancerous tissues.Methods:Collect breast cancer tissue and breast tissue adjacent to carcinoma 30 pairs respectively, Do not select the same quadrant, and normal tissue adjacent to carcinoma at least more than 5cm away from the edge of primary breast cancer tissue as mammary gland tissue adjacent to carcinoma. Respectively, extract total RNA, reverse transcription, the expression difference of MALAT1 in paired breast cancer, adjacent normal tissues were by the method of Real Time Polymerase Chain Reaction;Spin column extraction using breast cancer tissue and adjacent tissues of genomic DNA(gDNA),Using methylation-specific PCR (MSP) to detect gene expression differences MALAT1 methylation and breast cancer between adjacent groups of MALAT1 gene methylation in breast cancer were analyzed, all Statistics all analyses were performed with the statistical software spss 17.0. The results make a statistical processing and analysis.Results:1.Through detecting the fluorescent quantitative real-time PCR, the expression in 30 breast cancer tissues and the paired adjacent tissues,the expression quantity of MALAT1 in breast cancer tissue is 6.9±2.22, which is significantly higher than normal tissue adjacent to carcinoma (4.69±3.42), P=0.046;2. By MSP detection, there are 5 cases of breast cancer tissues appearing methylation of MALAT1, not methylated 25 cases; While Methylation in tissue adjacent to breast carcinoma with 12 case, and 18cases have no methylation,.P=0.04;Conclusions:1.This experiment found that the expression of MALAT1 in breast cancer tissue is significantly higher than normal tissue adjacent to breast cancer, difference has statistical significance, the expression level of MALAT1 are obviously Increase in breast cancer than other indicated that MALAT1 may be closely related to the development of breast cancer, bring into play the role of tumor suppressor genes.2.This experiment found that the expression of DNA methylation in breast cancer tissue is significantly less than normal tissue adjacent to breast cancer, difference has statistical significance,these findings indicate indicate that silencing of MALAT1 due to promoter hypermethylation is a frequent event and may contribute to Inhibit breast cancer development,MALATl may be a valuable biomarker for the study of breast cancer carcinogenesis and progression.
