Abstract/Summary:
Background: At present,MALAT1 gene polymorphism has become a hot spot of research.A large number of studies have been conducted that MALAT1 gene polymorphism is associated with the risk of neoplastic disease.However,the research on MALAT1 gene and coronary heart disease(CHD)is confined to MALAT1 molecular level,to our best knowledge,there is no report on MALAT1 gene polymorphism in the he risk of coronary artery disease.So we proposed a case-control study of 2 gene polymorphisms(theSNP of rs4102217 and rs619586)and coronary artery disease(CAD),to explore the relationship between MALAT1 gene polymorphisms and the risk and related factors.Objective:We aimed to explore the association of 2 polymorphisms rs619586(A/G)and rs4102217(C/G)in LncRNA MALAT1 gene with CAD risk and related factors.Methods:We collected a total of CAD patients(n=365)in Liaoning province and age and sex matched controls(n=384)at the same period.Genotyping of rs619586,and rs4102217 were performed using KASP method.Between-group differences in sex variability,as well as the Hardy Weinberg Equilibrium,were compared by the χ2 test and by analysis of variance for age variability.Multivariate logistic regression to analyze the odds ratio and 95% confidence interval with adjustments for age and sex The haplotype of each gene was analyzed using SHEsis software.was used to show the association between selected gene polymorphisms with CAD risk.When we analyzed the relationship of three SNPs and clinical features,if the dominant model for disease contribution P value is less than the recessive model,we selected the dominant model;othewise we seleted the recessive model.P value <0.05 was considered significant.Results:A total of 365 patients with coronary artery disease and 384 control people were involved in our study.Analyzing the essential characteristic of case group and control group,there were no statistic difference in demographic data such as age,gender,and triglyceride(P>0.05).Some risk factors are much higher in case group than contol group,such as smoking,high blood pressure,diabetes,cerebrovascular disease,hyperlipidemia.Drinking,high-total cholesterol and high low-density lipoprotein were higher in case group than in contol group,the difference was statisticallysignificant(P<0.05).MALAT1 rs4102217 was G/C variation in gene exon.there were three genotypes,respectively GG,GC and CC.no significant result was found in MALAT1rs4102217 polymorphisms between the risk factor of coronary artery disease in single factor analysis,we performed stratification analysis to eliminate other risk factors of coronary heart disease,the stratified analyses showed the MALAT1rs4102217 polymorphisms GC and CC+GC genotype promot the risk of CAD than those with the AA genotype in non-drinking subgroup(respectively P=0.010,OR=1.96,95%CI:1.17-3.28 or P=0.026,OR=1.73,95%CI:1.07-2.79).Diabetes subjects carrying GC and CC+GC genotype 4.07 times and 3.29 times increased risk of CAD than those with AA genotype(respectivelyP=0.010,OR=4.07,95%CI:1.41-11.81orP=0.019,OR=3.29,95%CI:1.22-8.88).MALAT1 rs619586 was A/G variation in promotor region.there were three genotypes,such as GG,GC and CC.no significant result was found in MALAT1rs619586 polymorphisms between the risk factor of coronary artery disease in single factor analysis.After adjustment by gender,age,smoking,drinking,high blood pressure,diabetes,low-density lipoprotein and odds ratio,also,no significant result was found in MALAT1rs619586 polymorphisms between the risk factor of coronary artery disease.Further analysis was conducted on the interaction between MALAT1 SNPs,the results of haplotype analysis indicated that there was no association between MALAT1rs4102217-rs619586 polymorphisms C-A,G-A,G-G haplotypes and coronary heart disease.Conclusion : Our study demonstrates that the rs4102217C/G polymorphisms of MALT1 are associated with the risk factors of CAD in non-drinking or diabetes subgroup in the Chinese population,and can to predict CAD risk.
