OBJECTIVE To investigate the relationship of three single nucleotide polymorphisms of ADP-ribosylation factor-like 15 (ARL15) gene and adiponectin gene with T2DM in Chinese-Korean in Yanbian.METHODS The project was designed as a case-control study. A total of 144 subjects with T2DM (T2DM, n=144), and 140 age-gender matched control subjects with normal glucose tolerance (NGT, n=140) were involved in this study. The genotypes of rs26770 (A/G) in ARL15 gene and rs 1501299 (C/A) and rs2241766 (T/G) in adiponectin gene were detected by the method of SnaPshotMultiplex. In the morning and fasting state, height, weight, waist circumference, hip circumference and blood pressure were measured and 5 ml of anti-coagulation peripheral blood was collected. The serum glucose (FPG), blood lipid (TC, TG, HDL-C, LDL-G) were detected and waist-to-hip ratio (WHR) were calculated. The plasmic ARL15 (PARL15), adiponectin (PA) and insulin (INS) were detected by using the enzyme linked immunosorbent assay.RESULTS 1. Genotypic distributions and allelic frequencies of rs26770, rs 1501299 and rs2241766 were compatible with the Hardy-Weinberg equilibrium and showed no significant difference (P>0.05) among every group.2. The frequency of The combined genotype GG-AA-TT of rs26770ã€rs1501299 and rs2241766 was higher in T2DM group than in NGT group and the difference had statistically significant (P<0.05).3. The level of PA in T2DM group was significantly lower than in NGT group, but the level of PARL15 was obviously higher than in NGT group. The differences had statistically significant (P<0.05).4. The level of PA in NGT group carrying the genotype TT of rs2241766 in adiponectin gene was significantly lower than the genotype GG+GT, but the level of TG was obviously higher. This inheritance showed dominant mode. The level of HDL-C and BMI in T2DM group having the genotype GG of rs26770 in ARL15 gene was lower than the genotype AA+AG, but the level of PA was higher. This inheritance showed recessive mode.5. Compareing the allele frequencies of rs26770, rs1501299 and rs22417663 in Chinese-Korean in yanbian with Han Chinese in Beijing (CHB), Western European ancestry from the CEPH collection (CEU), Japanese in Tokyo (JPT) and Yoruban in Ibadan (YRI) in HapMap database, the G allele frequency of rs26770 was obviously lower than CEU (P<0.01), but there was no significant differences with HCB JPT and YRI (P>0.05). The A allele frequency of rs1501299 was lower than YRI and had no differences with CEB, JPT and YRI. The G allele frequency of rs2241766 was significantly highly than CEU and YRI (P<0.01) and had no differences with CHB and JPT (P>0.05).CONCLUSIONS1. The combined genotype GG-AA-TT of rs26770, rs1501299 and rs2241766 in ARL15 gene and adiponectin gene in Chinene-Korean in yanbian increases the risk of developing T2DM.2. The genotype GG of rs2241766 in adiponectin gene up-regulates the level of PA and down-regulates the level of TG 3. The high plasmic ARL15 is highly associated with T2DM.4. There is a racial and ethnic difference in the polymorphisms of rs26770, rs1501299 and rs2241766 in ARL15 gene and Adiponectin gene. |
