| Objective:1. To study the effect of OSW-1 and deletion of mitochondrial DNA on PI3K signaling pathway in hepatoma carcinoma cell.Methods:1. Establish the mitochondrial DNA deletion model, p0-SK-Hep-1 (e.g.2), which was maked of the SK-Hep-1 cell(c.g.) as the control group. Compare the difference of expression between SK-Hep-1 andp0-SK-Hep-1 cell on PI3K signaling pathway by a quantitative real-time PCR Assay.2. SK-Hep-1 (e.g.1) and p0-SK-Hep-l(e.g.3) cell are intervened by OSW-124h, and compare the two groups’ different expression.Results:1. The up-regulated expression of experimental group 1 compares with control group:CASP9ã€CD1ã€FOXO1ã€GJA1ã€GRB2ã€HSPB1ã€IGF1ã€MAPK3〠PDPK1ã€PRKCAã€RPS6KA1ã€TLR4ã€TSC2. The down-regulated expression of experimental group 1 compares with control group:APCã€CTNNB1.2. the up-regulated expression of experimental group 2 compares with control group:FOS〠GRB2ã€HSPB1ã€IRAK1ã€PDPK1ã€PRKCAã€PRKCBã€TLR4ã€TOLLIPã€TSC2. The down-regulated expression of experimental group 2 compares with control group: APCã€IGF1ã€IRS1ã€PDGFRAã€PTPN11ã€SHC1.3. The up-regulated expression of experimental group 3 compares with control group:AKT1ã€CDC42ã€CDKN1B〠FASLBã€IRAK1ã€IRS1ã€PDGFRAã€PDK2ã€PIK3R2ã€PRKCBã€SHC1ã€TIRAP〠TOLLIP. The down-regulated expression of experimental group 3 compares with control group:BADã€EIF2AK2ã€FOXO1ã€IGF1ã€PABPC1ã€PAK1ã€PDK1ã€PDPK1〠PIK3CGã€RAC1ã€RASA1ã€RHOAã€SOS1ã€TLR4ã€WASL.4. The up-regulated expression of experimental group 1 and experimental group 3:IRAK1ã€PRKCB. The down-regulated expression of experimental group 1 and experimental group 3:IGF1〠APC.Conclusion:1. OSW-1 and deletion of mitochondrial DNA could effect the expression of PI3K signaling pathway in SK-Hep-1 cell.2. The targeting effects of OSW-1 on the mitochondria of SK-Hep-1 was proved.3. OSW-1 inhibited IGF-1-PI3K-AKT, Grb2-p85-AKT, IRS1-PI3K-AKT-GLUT-4, PI3K-AKT-TSC1/ TSC2- mTOR and TLR4-PI3K-AKT signaling pathway to inhibit hepatoma carcinoma cell’s growth, multiplication, energy metabolism. |
