Effects Of Acupuncture Inhibiting MMP-2, MMP-9, And Inflammatory Factor On Therapeutic Time Window Of Rat Cerebral Infarction

Objective:To observe the influence of acupuncture in regulating the activity of MMP-2, MMP-9, and the mRNA expression of inflammatory factor IL-1、 TNF-α、ICAM-1 on improved cerebral infarction rat model caused by autologous blood emboli.Methods:Male SD rats were randomly divided into six groups:normal group, model group, sham group, simple thrombolysis group, acupuncture combined with thrombolysis group, drug combined with thrombolysis group, among which, simple thrombolysis group, acupuncture combined with thrombolysis group and drug combined with thrombolysis group were divided into two subgroups:thrombolysis after 6 hours and after 9 hours after ischemia. Urokinase (UK) is intravenous thrombolytic drugs. Two hours after the successful establishment of model, the acupuncture combined with thrombolysis group received XNKQ acupuncture method, while the drug combined with thrombolysis group received matrix metallo-proteinase inhibitor treatment of doxycycline. Improved cerebral infarction model caused by autologous blood clot was used to evaluate neurological function according to Bederson to judge whether model establishment was successful. Took out the brain after 24 hours of successful model establishment, used the TTC staining method to calculate the infarction volume percent of each group.Real-time PCR was used to measure the mRNA expression of inflammatory factor TNF-α、ICAM-1. Gelatin-zymography is applied to measure the activity of MMP-2 and MMP-9 in the cerebral infarction of rats of each group.Results:(1)Cerebral infarction volume percent:Compared with the model group, it is not obvious of simple thrombolysis group(9h) to decrease the cerebral infarction volume, it was no significant difference(P>0.05), the cerebral infarction volume percent of simple thrombolysis group(6h), acupuncture combined with thrombolysis group(6h、9h), drug combined with thrombolysis group(6h、9h) was less than model group, it was statistically significant(P<0.01); the effect of acupuncture combined with thrombolysis group was better than simple thrombolysis group, it was statistically significant(P< 0.01); the effect of acupuncture combined with thrombolysis group was better than drug combined with thrombolysis group, the difference was significant(P<0.05).(2)The mRNA expression of IL-1、TNF-α、ICAM-1:Compared with model group, the mRNA expression of simple thrombolysis group(9h) was no significant difference(P> 0.05), simple thrombolysis group(6h), acupuncture combined with thrombolysis group(6h、9h), drug combined with thrombolysis group(6h、9h) could reduce the mRNA expression, the difference was statistically significant(P< 0.01);Compared with acupuncture combined with thrombolysis group, the effect of acupuncture combined with thrombolysis group was better than simple thrombolysis group, it was statistically significant(P<0.01); the effect of acupuncture combined with thrombolysis group was better than drug combined with thrombolysis group, the difference was significant(P<0,05).(3)The activity expression of MMP-2:Compared with model group, the activity expression of simple thrombolysis group(9h) was no significant difference(P> 0.05), simple thrombolysis group(6h), acupuncture combined with thrombolysis group(6h、9h), drug combined with thrombolysis group(6h、9h) could inhibit the activity expression of MMP-2, it was statistically significant(P<0.01); Compared with acupuncture combined with thrombolysis group(6h),the effect was better than simple thrombolysis group(6h) and drug combined with thrombolysis group(6h), it was significant(P<0.05); Compared with acupuncture combined with thrombolysis group(9h),the effect was better than simple thrombolysis group(9h), it was statistically significant(P<0.01),and the effect was better than drug combined with thrombolysis group(9h), the difference was significant(P<0.05).(4) The activity expression of MMP-9:Compared with model group, the activity expression of simple thrombolysis group(9h) was no significant difference(P> 0.05), simple thrombolysis group(6h), acupuncture combined with thrombolysis group(6h、9h), drug combined with thrombolysis group(6h、9h) could inhibit the activity expression of MMP-9, it was statistically significant(P<0.01); Compared with acupuncture combined with thrombolysis group, the effect was better than simple thrombolysis group, it was statistically significant(P<0.01),and the effect was better than drug combined with thrombolysis group, it was significant(P<0.05).Conclusion:Acupuncture could reduce the cerebral infarction volume, lower the mRNA expression of IL-1、THF-α、ICAM-1 and inhibit the activity of MMP-2、MMP-9.The effect was better than simple thrombolysis group and drug combined with thrombolysis group. It has a positive effect on prolonging the thrombolysis time window, and it provided new train of thought for future thrombolysis treatment of cerebral infarction.
