Xanthii Fructus first contained in "Shen Nong’s Herbal Classic", named Xi earshi; “Cang, erzi” first appeared in the "Qianjinshizhi", the calendar version of "Chinese Pharmacopoeia" collection contains. It is the dried ripe fruit with involucre of Xanthium sibiricum Patr. Xanthii Fructus has been provided with scatter cold, nasal passage, rheumatism of function.It was the traditional medicine to therapy nose as well as It has a long history of medicinal and Abundant resources in China.Xanthii Fructus of raw product is toxic,but it’s processing products was used in medicinal. Pharmacological experiments and clinical applications show that Xanthii Fructus of the toxicity was significantly reduced After processing. But it was not clear the part of Xanthii Fructus toxicity.Therefore, this paper uses the method of orthogonal experimental design, to preferred out the optimum extraction process and the optimum purification process. Further study on the compounds were isolated. By HPLC-MS methods were compared the chemical compoments of its toxic sites before and after processing, mainly in the following contents:1.Preferred out the optimum extraction process and the optimum purification procesGet toxic parts of Xanthii Fructus water extracts by D-101 macroporous resin,Orthogon experiment design a total of nine kinds to preferred out the optimum extraction process, the results showed that: useing water 10 times of Xanthii Fructus to extract 3 times,per time one hour was the optimal extraction process.Using D-101 macroporous resin for kaurene glycosides was the most applicable. Orthogon experiment design to preferred out the most applicable factoes of sample concentration, elution flow rate, height and diameter of the column, etc.The results showed that the optimum adsorption conditions for elution flow rate 1BV / h, the sample concentration of 1.0g crude drug / m L,height was 8times of diameter can get the maximum amount from the elution of macroporous resin; On the basis of the sample volume and the amount of eluent to inspect the results displayed on the sample volume: resin volume of 0.5: 1 when not appear on the sample fluid leak, the eluent was 4 column volumes can be the sample solution kaurene glycosides all eluted.And proof the process parameters by pilot magnification.2.Studies on quality standard of water extract of Xanthii FructusAccordance with the "Chinese Pharmacopoeia" Appendix collection contains inspection methods to check the moisture content of Toxic Parts. Using the method of single factor test, establish the process of colorimetric determination of kaureneglycosidecolorimetry. A comparative study of the content which about Xanthii Fructus included main active ingredient chlorogenic acid, 1,5- Dicaffeoylquinic acid and the main toxic components: carboxyl Atractyloside and Atractyloside by HPLC.3.Composition Analysis Research of the Toxic Parts of Xanthii FructusUsing silica gel column chromatography,combined SephadexLH-20 gel column chromatography, semi-preparative HPLC chromatography techniques to isolated and purified the kaurenoic acid from Xanthii Fructus, get two compounds: 2-O-2 [2-O-Isovaleryl-β-D-glucopyranosyl]-4-epi-atractyligenin(1) and β-daucosterol(2), wherein the compound(1) were isolated from Xanthii Fructus first time.4.Studies on chemical constituents of toxic sites of Xanthii Fructus and processing Fructus Xanthii by HPLC-MSFor the toxic parts in the first part,using HPLC-MS analysis of the technical means were detected out of 22 major component peak,By comparison of the two chromatograms found that a toxic constituent named 4’-desulphate-atractyloside content decreased significantly in the processing Fructus Xanthii.
