Objective: Based on the thought of TCM syndrome differentiation and treatme nt, this experiment probes into the influence of Qianggan Ruanjian Pills on the expr ession of liver fibrosis rat liver tissue MMP-1 and TIMP-1, expounds the therapeuti cal effect of Qianggan Ruanjian Pills’ anti-liver fibrosis by adopting modern animal experiment methods and establishing the liver fibrosis rat model induced by porcin e serum through Qianggan Ruanjian Pills, and provides reliable experimental basis f or the future further and extensive application of Qianggan Ruanjian Pills in the cli nical treatment of related liver fibrosis diseases and drugs.Methods: 120 SD male rats were selected in this experiment, weight of 180-200 g. Randomly divided into 6 groups:Normal control group, Model control group, Colc hicine intervention group, Colchicine treatment group, Qianggan Ruanjian Pill interven tion group, Qianggan Ruanjian Pill treatment group. Except the normal control group,each group use uninactivated porcine serum intraperitoneal injection to manufacturing models, two times a week, each time each one 0.5ml, 10 weeks. Normal control gr oup with the same method, the same dose, with the of intraperitoneal injection of no rmal saline. The intervention group was intragastricing administration at the same tim e intraperitoneally injecting, one time a day, with colchicine solution: 0.1g/Kg(colchi cine concentration 0.01g/ml), Qianggan Ruanjian Pill solution 2g/Kg(drug concentrati on 0.2g/ml). The normal group, the model group and the treatment group with the sa me dose of sterile water gavage every day. At the end of the fifth week, each group will select 5% rats randomly and kill them by decapitation method, then liver patho logy will prove that we make model successed. At the beginning of sixth week, each treatment group will given intragastric administration of drugs, and the same dose in each intervention group. Each group selected ten rats randomly and decapitated at th e end of the tenth week. Methods routine HE staining and Masson staining to observ e the morphological changes of liver tissue in rats of each group by cytology; immu nohistochemical staining, in liver tissue were detected by MMP1 in each group of m atrix metalloproteinase and its inhibitor TIMP1 using image analysis.Results: ① General conditions of rat livers in each group: normal control gr oup: little dark red, finely-rounded edge, smooth and unspotted surface; model contr ol group, dark red, finely-rounded edge, and the surfaces of most rat livers have bu lges; the livers of rats in the Qianggan Ruanjian Pill group and the colchicine grou p grow; the edges of a few rat livers is finely-rounded; dot bulges are found in the surface, but no significant changes in the pathological model group; ② Through o bservation over HE staining and Masson staining, it is found that Qianggan Ruanji an Pills are able to improve the extent of liver fibrosis of immunity-damaged liver fibrosis rats; ③ Compared with the normal control group, the content of the model control group, the colchicine group and the Qianggan Ruanjian Pill group is increa sed and has prominent statistical differences(P<0.05); compared with the normal co ntrol group, the expression of the colchicine group and the Qianggan Ruanjian Pill group is increased and has statistical meaning(P<0.05), which indicates that Qiangg an Ruanjian Pills can improve the expression of Matrix Metalloproteinase MMP-1; ④ Compared with the normal control group, the content of the model control grou p, the colchicine group and the Qianggan Ruanjian Pill group is decreased and has prominent statistical differences(P<0.05); compared with the normal control group, t he expression of the colchicine group and the Qianggan Ruanjian Pill group is decr eased and has prominent statistical differences(P<0.05); Qianggan Ruanjian Pills ca n reduce the expression of TIMP-1;Conclusion: Qianggan Ruanjian Pill is able to improve the expression of Ma trix Metalloproteinase MMP-1, reduce the expression of TIMP-1, thereby inhibit the liver fibrosis pathological change and produce the effect of anti-liver fibrosis.
