Effects Of Qiliqiangxin Capsule On The Renal Blood Flow, Oxidative Stress, Inflammatory Reaction On Acute Cardiorenal Syndrome In Rats

Objective :To discuss the effect of Qiliqiangxin capsule on the pathogenesis of acute cardiorenal syndrome, study the intervention of Qiliqiangxin capsule for the renal blood flow, decrease oxidative stress, inflammatory reaction in rats.Methods: Healthy SD rats 80,age:8 weeks, Weight: 200±20g, provided by the Animal Experiment Center of Henan province. The acute cardiorenal syndrome model preparation in two steps.The rat was anesthetized with 10% chloral hydrate(3ml/kg) by intraperitoneal injection, removed the right kindey trouhg the back incision; After 1 week,ligated the coronary artery witch between pulmonary arterial cone and auricula sinistra with 6.0 silk. All the rats application of the ultrasonic examination check the heart function before the surgery.Intraoperative electrocardiogram monitoring.Collected urine after 2h, 4h, 6h, 8h by metabolic cage and detected the NGAL. All groups ultrasonic examination to check the heart function 24 h after the MI sugery,and after 24 hours of myocardial infarction,Qiliqiangxin capsule high,medium and low group was divided according to 1.2 g/kg, 0.6 g/kg, 0.3 g/kg of 3 doses add distilled water 3 ml, mixture solution, filled with filling and feeding needle feed, for four weeks. Detected the renal blood flow by Sinmens Acuson Sequoia ultrasound machines, 15l8-s probe, the frequency of 10 MHZ, mechanical index was 0.18, the sampling volume of 2.0 mm, wall filter 100 hz, blood flow and the sound velocity Angle of < 30°, measured peak blood flow velocity(Vmax), end-diastolic velocity(Vmin), mean blood flow velocity(VM), renal blood flow resistance index(K-PI) formula: K-PI = Vmax- Vmin/VM. Four weeks later, each rat was executed by takeing blood, detected the biochemical indexes like parallel oxidative stress index(MDA, SOD), inflammatory factor index(TNF-a, IL- 10).Results : All the results of rats’ heart ultrasound examination before the sugery:compared to the sham group with the building model group on LVEF、LVFS、CO、LVDs、LVDd and LVESV,there was not any statistically significant difference(P>0.05);The operation group ST segment increased significantly; Building scores compared with control group after each period(2 h, 4 h, 6 h, 12 h) of the testing results of urine NGAL was increased significantly(p < 0.01); SOD in model group were significantly lower than the control group(P < 0.01), Qiliqiangxin capsule each dose group compared with model group expression of SOD increased significantly(P< 0.05); The MDA express of control group was significantly lower than the model group(P< 0.01), Qiliqiangxin capsule each dose group was significantly lower than the model group(P < 0.05); The model group IL-10 expression lower than the control(P < 0.01), Qiliqiangxin capsule each dose group of IL-10 compared with model group was significantly increased(P<0.01); The TNF-a expression of control group was significantly lower than the model group(P < 0.01), Qiliqiangxin capsule each dose group of TNF-a compared with model group was significantly decreased(P<0.05). Qiliqiangxin capsule each dose group of renal blood flow resistance index was significantly lower than the model group, renal artery blood flow resistance index fell by 7%, 18% and 30%, respectively, the difference between each group was obvious statistical significant(P < 0.01).Conclusion:(1)Qiliqiangxin capsule can improve the rats of acute cardiorenal syndrome of myocardial infarction renal blood flow, and decrease renal artery blood flow resistance index.(2) Qiliqiangxin capsule capsule can resistant the develop of rats’ acute cardiorenal syndrome of myocardial infarction through the mechanisms of inhibite oxidative stress and decrease the inflammatory reaction.
