| Objective:To confirm the impact of the Yishen Jiedu Tongluo capsule on the expression of Smad2 protein and the pathological morphology and structure diabetes mellitus rats.Methods:60 male SD rats,10 rats were treat as lank group,and the oher 50 rats were treated with intraperitoneal injection of STZ to induce diabetes mellitus model and then randomly divided into 5 groups:normal control group,model group,high dose treatment group,middle dose treatment group and low dose treatment group,and each group has 10 rats.All groups were given Yishejieduntongluo capsule suspension gavageand continuous adiabetes mellitusinistration for 12 weeks,then observe the general situation of rats,to detect the expression of the activity of renal tissue Smad2 protein, kidney pathological morphology observed under light microscope and electron microscope.Result:1.Yishen Jiedu Tongluo capsule can effectively improve the body weight of rats in the model group decreased,food intake,water intake,urine volume increased gradually such as the general situation.2.Each treatment group can significantly reduce the positive expression in rat glomerular mesangial region Smad2(P< 0.05, P<0.01),the positive expression of at least in which the Chinese medicine high dose treatment group,(P<0.01).3.All The treatment groups can improve the renal pathological changes such as model group appear:changes in extracellular matrix accumulation,glomerular basement membrane thickening,pyknosis,fibrosis pathology,in which the Chinese medicine high dose treatment group improved the most obvious lesions lighter than other group.4.Compared with the model group,the dose in Chinese medicine treatment group and the control group was GBMT,PFPT of rat kidney was significantly reduced(P<0.05,P<0.01);high dose of Chinese medicine treatment group decreased the GBMT and PFPT were significantly better than the control group(P< 0.01).Conclusion:1.Yishen Jiedu Tongluo capsule can effectively reduce the expression of Smad2 protein in ratkidney DN activity,inhibition of renal fibrosis,and thus to protect the kidney function.2.Yishen Jiedu Tongluocapsule can improve DN rat renal pathological morphologicalstructures such the accumulation of extracellular matrix and basement membrane thickening and fusion of foot process,there by protecting the kidney function. |
