ObjectiveUnder the guidance of the basic theory of traditional Chinese medicine,the clinical application of Astragalus Mongholicus Injection preventive and treatment of patients with Acute Radiation-induced Lung Injury. Observing the pathological changes in lung tissue, examining the m RNA expression of CCL2 and the CCL2 protein content in lung tissue;testing the number of monocyte(CD68) in lung tissue by building the rat model of acute radiation-induced lung injury. Methods64 female Wistar rats were randomly divided into control group,irradiation group, irradiation plus Astragalus mongholicus injection group. In addition to the control group,a single dose of 20 Gy were given to build the rat model of acute radiation-induced lung injury. After rats being sacrificed at 1,2,4,8week,HE slice was used to observe the pathological changes in lung tissue, RT-PCR was taken to examine the m RNA expression of CCL2 and ELISA was applied to detect the CCL2 protein content in lung tissue;the number of monocyte(CD68) in lung tissue were counted by immunohistochem- ical assay. All data was used SPSS 19.0 statistical analysis software for analysis. All measurement data using the mean± standard deviation(x±s),between the two groups using t test, count data using c2 test. Difference was statistically significant at p value<0.05. Results1. The occurrence of acute radiation-induced lung injury: Observing the pathological changes in lung tissue. In the control group, the structure of alveolar tissue is clear and we can not see inflammatory cell in lung issue. After a week of radiotherapy, we can see a little of inflammatory cells in lung issue. After two weeks of radiotherapy, we can see some inflammatory cells in lung issue and pulmonary edema. After four weeks of radiotherapy, become severe pulmonary edema and a large number of inflammatory cells in lung issue. After eight weeks of radiotherapy, the inflammatory is a little better.2. The changes of the CCL2 m RNA and the CCL2 protein in lung tissue: The relative m RNA expression of CCL2 in the control group,irradiation group,irradiation plus Astragalus mongholicus injection low dose group, irradiation plus Astragalus mongholicus injection dose group were(1.000±0.284),(6.983±0.576),(4.130±0.396),(2.975±0.413);the CCL2 protein content in lung tissue were(1.00±0.04),(3.52±0.11),(1.93±0.07)(1.18±0.05). The difference was significant(P < 0.05). 3. the number of monocyte(CD68) in lung tissue: the monocytes at high magnification field in each group were(10.0±2.4),(46.0±4.8),(27.0±3.5),(18.0±2.8), the difference was statistically significance(P<0.05). Conclusions1. Astragalus mongholicus injection significantly alleviated the lung inflammation in rats with acute radiation-induced lung injury.2. Astragalus mongholicus injection significantly decreased the m RNA expression of CCL2 and the CCL2 protein content in lung tissue and sera.3. Astragalus mongholicus injection inhibited the accumulation monocyte(CD68) in lung tissue4. There is a certain relation between the effect and the dose of Astragalus Injection on acute radiation lung injury.
