Objective To study the function of Chinese materia medica preparation Qingkailing injectiong and astragalus membranous Injection to acute lung injury(ALI)induced by lipopolysacCharide,and discuss their mechanisns.Method Eslablish animal model of acute lung injury.Forty Sprague-Dawley male mice are divided into five groups: normal group, model group ,astragalus membranous group, Qingkailing group and combined group.Show caudal vein up and inject medicaments from caudal veins of mice. Normal group:Inject 1ml saline from the caudal vein of mice. Model group, astragalus membranous group, Qingkailing group and combined group: Iinject lipopolysaccharide(5mg/kg) which dissolved in 1ml saline from caudal vein of mice;After forty-five minutes later, astragalus membranous group: Inject astragalus membranous injection (0.5g/kg) which diluted in 1ml saline from caudal vein of mice; Qingkailing group: Inject Qingkailing injection (0.05ml/kg) which diluted in 1ml saline from caudal vein of mice; combined group: Inject astragalus membranous injection (0.5g/kg) which diluted in 0.5ml saline and Qingkailing injection (0.05ml/kg) which diluted in 0.5ml saline from caudal vein of mice. Five hours later from the first injection,take anticoagulation blood (1:500u heparin sodium sodium heparin )out of abdominal aorta and do blood gas analyse immediately.Then take two milliliter anticoagulation blood out of abdominal aorta to do flow cytometry.Last, put mice to death.with flowing blood up. Observe lung tissue by visual inspection and×200 microscope. Weighing the right-down-lung wet weights,dye weights,rate of wet weights to dye weights of this mice. The expression of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β)in the lung tissue was detected by immunhistochemical method , in pathology image analysis system determination TNF-αand IL-1βaverage grey. Measure the number of CD4 +cells,CD8+cells and the rate of CD4 +cells to CD8+cells (CD4+/CD8+)with flow cytometry.Results None of forty mice in this study are lost or die.And all of the results are in the data analytics.①Observe lung tissue by visual inspection:The surfaces of normal group's lung tissue are slick,rose pink,no congestion,no dropsy and no infarct. The surfaces of model group's lung tissue are wine, congestion, dropsy and many petechia. Astragalus membranous group'lung tissue surfaces are some congestion, dropsy and little petechia,but less than normal group's. Qingkailing group and combined group are the same as Astragalus membranous group'. Observe lung tissue by×200 microscope:The structures of normal group's lung tissue are clear, no congestion ,no effusion,no inflammatory cell soakage. model group's lung tissues are congestion, dropsy and tiny thrombosis,and there are abundant of edematous fluid and inflammatory cells soakage in lung stroma and alveolus,and some lung tissue can be see punctation pulmonary atelectasis.The lung tissue of astragalus membranous group are congestion, dropsy ,tiny thrombosis and inflammatory cell soakage,but it is slighter than model group's; Qingkailing group and combined group are the same as astragalus membranous group'.②The right-down-lung wet weights,dye weights,rate of wet weights to dye weights of normal group mice are less than any other groups'(P﹤0.05). The right-down-lung wet weights,dye weights,rate of wet weights to dye weights of model group mice are more than any other groups(`P﹤0.05).There is little difference between astragalus membranous group and Qingkailing group;The dye weights of combined group are lighter than Qingkailing group'(P﹤0.05).There are no difference between Qingkailing group's wet weights,rate of wet weights to dye weights and combined group's. There are no difference between astragalus membranous group's wet weights,dye weights,rate of wet weights to dye weights and combined group's.③All of groups'PaCO2 are little differences(P>0.05). Normal group`s pH,PaO2,PaO2/FiO2 is more difficult than other groups('P﹤0.01),and pH is heightening,then PaO2 and PaO2/FiO2 is letdown.Model group's PaO2 and PaO2/FiO2 is less than any other groups'(P﹤0.05), especially combined group's(P﹤0.01).Each of astragalus membranous group,Qingkailing group and combined group is no differences in pH,PaO2,PaO2/FiO2.④The expression of TNF-αand IL-1βof normal group is more remarkable difference than any other groups',and The expression of TNF-αand IL-1βof normal group is the least in all of groups. The expression of TNF-αand IL-1βof model group is the highest in all of groups and is more remarkable difference than any other groups'(P﹤0.05). The expression of TNF-αand IL-1βof combined group is less than astragalus membranous group's and Qingkailing group's,and there are remarkable differences(P﹤0.05).⑤There is remarkable distinction only between normal group and model group about the number of CD8+ cell(P﹤0.05),but there is no difference in other groups. The number of CD4 +cell of normal group is more than any other groups',and the differences are remarkable(P﹤0.05). The number of CD4 +cell of model group is less than any other groups',and the differences are remarkable(P﹤0.05). The number of CD4 +cell of combined group is more than astragalus membranous group's and Qingkailing group's. There are no remarkable differences between CD4+/ CD8+ of combined group and normal group`s. normal group's CD4+/ CD8+ and combined group's CD4+/ CD8+ are higher than astragalus membranous group's and Qingkailing group's (P﹤0.05).But model group's CD4+/ CD8+ is less than astragalus membranous group`s and Qingkailing group's (P﹤0.05).conclusion ①Both Qingkailing injectiong and astragalus membranous injection can lighten pulmonary edema;②Both Qingkailing injectiong and astragalus membranous injection can ameliorate gas exchange in ALI,and slower degree of respiratory distress;③Both Qingkailing injectiong and astragalus membranous injection can Inhibit the expressions of inflammatory factor as TNF-αand IL-1β;④Both Qingkailing injectiong and astragalus membranous injection can highten the number of CD4+ and adjust CD4+/ CD8+;⑤Qingkailing injectiong combination with astragalus membranous injection could produce synergetic effect.
