The Research On The Attenuation,Anti-Inflammatory And Analgesic Effects Of Guizhi Decoction To Strychni Through The Animal Experiment

Objective:To provide the exper imental basis for the clinical application of nux vomica.First, measure the t oxicity of nux vom ica, then m easure the toxicity of Nux vomicaof different dose compatibility of Baishao Guizhi Decoction, Chishao Guizhi Decoction and Chi baishao Guizhi Decoction Re spectively, lastresearch t he effect of Guizhi Decoction which can reduce the toxicity of nux vomic athat on anti-inflammatory and analgesic effects of nux vomicathrough the animal experiments.Methods:(1)D etect the toxicity of nux vomica through the Bliss method,using the NDST software calculate the results.(2)Using different dose of Nu x vomica comp atibility of Baishao Guizhi Decoction, Chishao Guizhi Decoction and C hibaishao Guizhi Decoction Respectively, measure the LD50 r espectively.(3) Through the ani mal experiments, can get a Guizhi Decoction which can reduce the toxicity of nux vomica, through the capillary permeability test and ho t plate test experiment s to study its eff ects on anti-inflammatory and analgesic effects of nux vomica respectively.Results:(1)Through animal experiment, get the prepared nux vomi ca LD50 i s 258.07mg/kg.(2)The toxicity of the different dose groups of Baishao Guizhi Decoction compatibility of nux vomica, that is the group of Baishao Guizhi Decoction compatibility of 13.41% nux vomica, the group of Baishao Guizhi Decoction compatibility of 7.19% nux vomica, the group of Baishao Guizhi Decoction compatibility of 3.72%nux vomica, the LD50 are 8218.21mg/kg, 3486mg/kg, 1642.586mg/kg respectively. T he LD50 of the same proportion of Chishao Guizhi Decoction co mpatibility of nux vomic a are 9335.06mg/kg, 3591.47mg/kg, 2064.06mg/kg. T he LD50 of t he same proportion of Chiba ishao Guizhi Decoction compatibility of nux vomica are 8228.82mg/kg, 2972.45mg/kg, 1792.80mg/kg. According to the detoxification rate =(after the compatibility of the LD50-Nux Vomica LD50) / Nux Vomica LD50number*100%, We can calculate the detoxi fication rate of different group: The detoxi fication rate of Baishao Guizhi Decoction compatibility of 3.72% nux vom ica is 18.5%; The detoxif ication rate of Chis hao Guizhi Decoction compatibility of 13.41 % nux vomica is 7.28%; The detoxification rate of Chishao Guizhi Decoction compatibility of 7. 19% nux vomica is 0.08%; The detoxi fication rate of Chishao Guizhi Decoction compatibility of 3.72% nu x vomica is 34.6%; The detoxification rate of Chi-baishao Guizhi Decoction compatibility of 3.72% nux vomica is 18.6%. The experimental results show that three di fferent group of Chishao Guizhi Decoction compatibility have ou tstanding effect on reducing poison of nux vomica, but the effection of reducing po ison has not obvious relati onship between compatibility proportion.(3)Through the experiment of capillary permeability in mouse, All the group of Chishao Guizhi Decoction(in Low hi gh school) compared with the group of nux vomica can reduce absorbance of celiac capillary in mouse, which effect of 13.41% nux vomica group significantly(P < 0.05), According to the Comparison each compatibility group capillary permeability in mouse, it is negatively correlated with the proportion of nux vomica.(4) Through the experiment of hot plate test experiments, All the group of C hishao Guizhi Decoction(in Low hi gh school) can not strengthen clearly effection on analgesic effect s of nux vom ica,but have Some synergistic effection on analgesic effects of nux vomica.Conclusion:Through the experimental studies of acute t oxicity and pharmacodynamics, comprehensive analysis show:Chishao Guizhi Decoction can detoxificate clearly on nux vomica,and strengthen effection on anti inflammatory and analgesic eff ects of nux vomica.
