Stroke is one of the diseases that threaten the health of the elderly and with high disabled rate, fatality rate, and many complications. And the ischemic stroke is the most common. TCM theory of the pathological mechanism of the ischemic stroke is Qi deficiency and blood stasis. Buyang huanwu decoction(BYHWD) is originate from "Yi Lin Gai Cuo" by great physician Wang Qingren in Qing Dynasty; It is the typical prescription for treatment of the ischemic stroke, and its primary efficacy is supplementing qi and activating blood circulation. The effective constituents in BYHWD are astragaloside Ⅳ and hydroxysafflor yellow A. Therefore, for the purpose of studying the material basis of drug efficiency of BYHWD in treatment of ischemic stroke, selected BYHWD formula granules as the object of study, used serum pharmacochemistry methods, and measured concentration changes of the effective constituents of BYHWD in animals, and provided references for illustrating the pharmacodynamic material basis of " invigorating qi and activating blood circulation" efficacy of BYHWD.Objective: 1. To establish a convenient and reliable method for content determination of the astragaloside Ⅳ and hydroxysafflor yellow A. And measure the concentration of the effective constituents in BYHWD formula granule and simplex drug with in vivo metabolism of rats, respectively.2. To compare the content change of the effective components of astragaloside Ⅳ and hydroxysafflor yellow A with and without metabolism in rats. And to determine the influence of BYHWD combination on its efficacy, and to interpret the material basis of the efficacy of BYHWD on supplementing qi and promoting blood circulation.Methods:Determination of the content of the astragaloside Ⅳ in BYHWD and study of pharmacokinetics in rats1 Content determination of the astragaloside Ⅳ in BYHWD formula granule and simplex drugHPLC-ELSD method was applied on content determination of the astragaloside Ⅳ in BYHWD formula granules and simplex drug Astragalus Membranaceus. By the optimization of the chromatographic conditions, sample preparation and pretreatment method, and methodology, the feasibility, the precision, the accuracy and reproducibility of this method was verified.2 Determination of drug concentration with metabolism and comparison of the content changes.rats were randomly divided into BWHWD formula granule group and Astragalus group, and was administered BYHWD solution and Astragalus Decoction(10ml/kg of weight) intragastrically for 8days, and then the serum and cerebrospinal fluid samples was collected; on the other, rats was administered intragastrically for 3 days,and transferred into metabolic cages, urine and fecal samples were collected respectively. Drug concentrations with and without metabolism was compared, relationship between content changes of the astragaloside Ⅳ and the material basis of BYHWD on supplementing qi was discussed.Determination of the content of the hydroxysafflor yellow A in BYHWD and study of pharmacokinetics in rats1 Content determination of the hydroxysafflor yellow A in BYHWD formula granule and simplex drugHPLC-UV method was applied on content determination of the hydroxysafflor yellow A in BYHWD formula granules and simplex drug Carthamus Tinctorius. By the optimization of the chromatographic conditions, sample preparation and pretreatment method, and methodology, the feasibility, the precision, the accuracy and reproducibility of this method was verified.2 Determination of drug concentration with metabolism and comparison of the content changes.The method of this part of the experiment was the same as that of the astragaloside Ⅳ.Results:1 The assay method in this experiment was in good linear correlation, retrieving rate, precision and stability, it was shown that this method has the advantages of simple operation, high sensitivity and little interference of impurity.2 The content of astragaloside Ⅳ and hydroxysafflor yellow A in cerebrospinal fluid,urine and fecal samples was very low, the quantitative analysis of content changes of the effective constituents with metabolism was difficult to perform.3 The content of astragaloside Ⅳ in BYHWD serum samples was 10% of the content in BYHWD solution, and the content of astragaloside Ⅳ in astragalus simplex drug serum samples was 4% of the content in astragalus decoction. And the content change of effective constituents in BYHWD granules into the blood was higher than that of the astragalus simplex drug.4 The content of hydroxysafflor yellow A in BYHWD serum samples was 1.2% of the content in BYHWD solution, and the content of hydroxysafflor yellow A in safflower simplex drug serum samples was 0.6% of the content in safflower decoction. And the content change of effective constituents in BYHWD granules into the blood was higher than that of the safflower simplex drug.Conclusion:The content change of astragaloside Ⅳand hydroxysafflor yellow A in concerted application of prescription BYHWD into the blood was substantially increased, compare to that of simplex drug astragalus and safflower. And it is indicated that concerted application of prescription take a promoting effect on supplementing qi efficacy of astragalus and activating blood circulation efficacy of safflower. And it provided a theoretical basis for clarification of the efficacy of BYHWD on invigorating qi and activating blood circulation.
