Healing Effect Of Blumea Balsamifera Oil On Deep Second Degree Skin Burn In Rats

ObjectiveBurn injury is frequently occurring disease in clinical, which is one of the most common and devastating forms of trauma, particularly partial and full-thickness burn. To investigate the healing efficacy of Blumea balsamifera oil(BB oil) in burn injury, the deep second degree burn rats were selected. Different tissues from overall to partial were detected, and elucidated the treatment effect and mechanism by BB oil.MethodsAll animals were randomly assigned to 9 groups of 24 each, including control group, 80% ethanol group(vehicle), burn ointment group(standard), 100% BB oil group, BB oil 50% group, 10% BB oil group, L-borneol group, D-camphor group and L-borneol + D-camphor group. Briefly, on the basis of other researches, the deep second degree burn model was established with an iron rod immersed in boiling water. After 1 h, all the animals were administered treatment and applied once daily, last for 21 days. The rats of weight, the rate of healing, scabs dropped time and re-epithelization time were observed every three days, during 21 days after burn injury. The samples were removed from different treated rats by sacrificing the 4 animals on the 1st, 2nd, 5th, 9th, 14 th, 21 st day post-burn established. Then the water content of burn tissue was measured. Plasma IL-1 and TNF-α levels were evaluated, and the tissue expressions of bFGF, VEGF, TGF-β, hydroxyproline were determined along with skin histopathology. After the analysis of information in the therapeutic process, clarity the treatment effect and mechanism by BB oil.ResultsThe general observation results showed that BB oil significantly reduced the water content of tissue, shortened the scabs dropped time and accelerated healing in the burn injury animals. And the activity of SOD and expressions of bFGF, VEGF, TGF-β and hydroxyproline significantly increased in the plasma or tissue, however, the levels of MDA, IL-1 and TNF-α on plasma decreased. Meanwhile, the results indicated that L-borneol and D-camphor particularly decreased the levels of MDA in plasma at the early stage of burn, and D-camphor particularly decreased the levels of TNF-α at the same time. All the indexes in the tissue showed that BB oil could increase the expressions of b FGF, VEGF, TGF-β and hydroxyproline in the tissue, further explained by the skin histopathology.ConclusionThis study showed that the BB oil facilitated wound healing in experimental animal model by influencing the antioxidant, anti-inflammatory, the expressions of cytokines and growth factors in burn injury repair process. A series of experiments on rats was carried out to determine the dominant compounds that are responsible for its biological activities in BB oil. Either L-borneol or D-camphor has effected on wound healing process, but not total effect. However, the results of L-borneol and D-camphor groups demonstrated that they has good effect on antioxidant and anti-inflammatory in animals, and they maybe the active ingredients in the BB oil for antioxidant and anti-inflammatory. Here is a need for further studies in order to analyze other active ingredients in the BB oil and to completely elucidate the mechanisms of actions of these active ingredients.
