Effect Of YangXueQingNaoKeLi On Synaptophysin And Microtubule-associated Protein 2 In CA1 Area Of Hippocampus Of Vascular Dementia Rats

Objective By observing the expression of synaptophysin and microtubule-associated protein2 in CA1 area of hippocampus of vascular dementia rats,and the influence of Yang Xue Qing Nao Ke Li on them, explore the influence of Yang Xue Qing Nao Ke Li on Vascular Dementia rats and the effect of synaptic plasticity.Methods 1 90 healthy male SD rats were randomly divided into sham operation group, vascular dementia model group and medicine treatment group. Each group was divided randomly into 1, 2, 4, 8 and 12 W 5 subgroups after the success of model preparation(n=6). 2 The topic took improved four vessles to block the rule of law as a vascular dementia model. 3 After the success of the model of preparation, the rats were given Yangxueqingnaokeli 3.2g/kg orally in medicine treatment group. 4 Testing the behavior of rats by Morris water maze method after 1, 2, 4, 8 and 12 W,and then processing in the brain tissue in rats. 5 We observe the pathological and histological changes in rat hippocampal CA1 area by HE staining and the changes of the ultrastructure of rat hippocampal CA1 area synapses. 6 Using immunohistochemical method measuring the rat hippocampal CA1 area SYN and MAP-2, and then using an electron microscope observation, finally using MOTAC pathological image system is analyzed. 7 Application SPSS 17.0 statistical software for data analysis at last. Measurement data are presented as mean±SD( sx ±),using anova design analysis of variance within the group, P<0.05 was statistically significant.Results 1 Groups of rats with the increase of training days, the average escape latency period are reduced, the previous two days, the rats did not change the average escape latency(P>0.05), the difference was not statistically significant. Starting from the first three days, compared with the control group, the model group average escape latency was significantly longer(P<0.05),the difference was kept on the fifth day. Compared with the VDgroup, the MTgroup were significantly shorter average escape latency(P<0.05), the first five days of the more significant differences(P<0.01), with statistical significance.After the withdrawal of security platform, compared with the control group, the VD group were significantly reduce the number of cross platform(P<0.05), Compared with VD group, the MT group passing through the platform number of times increased remarkablely(P<0.05),the difference was significant.2 Control group lined up in the rat hippocampus CA1 area of nerve cells, number of neurons, hippocampus nerve cells in the VD group were poorly arranged, significantly reducing the number of neurons, showing nuclear condensation,nuclear fragmentation and nuclear fusion phenomenon, the MT group in the rat hippocampus nerve cells compared with model group had obvious improvement, cell arrangement more closely, the number of neurons, earlier nuclear pyknosis, karyorrhexis phenomenon, reduce earlier.3 Under electron microscope, hippocampal CA1 neurons in cell membranes and nuclear structure intact control rats, abundant mitochondria, evenly distributed chromatin, clear boundary around the synaptic membrane, a complete outline of synaptic structure is clearly visible. The VD group rats hippocampus neuron cell membrane and nucleus structure owes clear, visible mitochondria expansion even burst, significant quantity reduce the cavitation status, presynaptic membrane and postsynaptic membrane damage, the synaptic cleft. The MT group in the rat hippocampus neuron damage is improved compared with the VD group, the cell membrane and nucleus structure is complete, the number of mitochondria is more than before, synaptic membrane before and after the border and clear, the smaller the synaptic cleft. 4 We can see more quantity of the SYN and the expression of the MAP-2 positive cells in control group at each time point, the difference was not significant(P>0.05).Compared with control group, VD group at each point in CA1 region of SYN、MAP-2 protein expression decreased,and the difference was valuble(P<0.05).Compared with VD group, the MT group at each point in CA1 region of SYN、MAP-2 protein expression increased,and the difference was significant(P<0.05). The expression slightly increased in 1W, reached peak at each time point in 4W, and slightly decreased in 8W,the expression is still high in12 W.Conclusions 1 Yang Xue Qingnao Ke Li is in favor of synaptic remodeling and can significantly improve vascular dementia rats learning and memory ability. 2 Yang Xue Qingnao Ke Li has therapeutic effect on vascular dementia, its function may be relevant with the increased expression of SYN and MAP-2 in CA1 area of hippocampus.
