Introduction:Currently, the diabetes has become the third serious chronic and non-infection disease. According to the modern pharmacological studies have shown that the fenugreek could low blood sugar, blood lipids and antioxidant-effect, which also could improve and regulate the complications of diabetes.The article combined the pharmacological effects, pesticide effects and chemical studies to determine the material base, thus including the polysaccharides, the saponins and the flavonoids. The article initially established the process of extaction and a set of relatively complete methodology about the Diosgenin and Orientin, which is based with “2010 edition Pharmacopoeia”.Objective:1. With the content of polysaccharides, saponins, flavonoids to establish the best process of extaction.2. With HPLC-ELSD technology to build the Diosgenin(polysaccharides) analytical approach and formulate the quality standards in the extraction.3. With HPLC-UV technology to build the Orientin(saponins) analytical approach and formulate the quality standards in the extraction.Methods:Firstly, with the content of polysaccharides, saponins, flavonoids as an indicator, Compare the reflux extraction, ultrasonic extraction, dipping extraction and percolation extraction, thus selecting the percolation extraction as the best method; Secondly, do the single factor to the process of extraction,including : solvent, concentration, granularity,time, solid-liquid ratio, solvent volume, velocity and times;Thirdly, the relative density Investigation; Lastly, study the extraction solvent, volume and times. In order to determine the optimum extraction processThough the mobile phase and the temperature drift tube to determine the optimal chromatographic conditions. According to the“2010 edition Pharmacopoeia”, the article has established the complete methodology—Diosgenin by HPLC-ELSD, including: special properties, sample processing methods(sample extraction method, hydrolysis-acid concentration, hydrolysis time, extraction volume and times),a linear relationship, system suitability, accuracy(repeatability, intermediate precision), accuracy and durability(sample weight, the proportion of the mobile phase, flow rate, injection volume, column type, column oven, the drift tube temperature, solution stability).etc, which is to establish the Diosgenin determination methodology and formulate the content of Diosgenin in the extraction.According to the“2010 edition Pharmacopoeia”, the article has established the complete methodology— Orientin by HPLC-UV, including: special properties, sample processing methods(sample extraction method, the extraction solvent, solvent ratio, extraction time, extraction times), a linear relationship, system suitability, accuracy(repeatability, intermediate precision), accuracy and durability(sample weight, the proportion of the mobile phase, PH value, flow rate, injection volume, column type, different chromatography, column oven, the drift tube temperature, solution stability).etc, which is build the Orientin determination methodology and formulate the content of Orientin in the extraction.Results:With the content of polysaccharides, saponins, flavonoids as an indicator and the single factor experiment to establish the optimum extraction process and conditions, Which is over 50 purpose fenugreek powder, eight times the volume of 60%Methanol, dipping 24 hours, 0.15ml/min of the flow rate, collecting 10 volumes the percolate. at 60 ℃ evaporated and concentrated to the relative density of 1.10 to 1.15, and then add ethyl acetate to extract(1/2 volume, twice).Under the selected chromatographic conditions, analyzing the content of diosgenin in the extraction, the conclusion is that the content of diosgenin could get(1.221 ± 0.010)%.According to the results to fall 20%, each gram of extract containing diosgenin shall not be less than 0.978%, thus building the standard of diosgenin.Under the selected chromatographic conditions, analyzing the content of Orientin in the extraction, the conclusion is that the content of Orientin could get( 1.055 ±0.184)%.According to the results to fall 30%, each gram of extract containing Orientin shall not be less than0.740%, thus building the standard of Orientin.Conclusion:In this paper, with the content of polysaccharides, saponins, flavonoids as an indicator, we initially established fenugreek optimum extraction process. Through the establishment of Diosgenin and Orientin analytical approach, we also initially established the quality of standard of saponins and flavonoids.
