Objective :After injecting D- galactose made the aging model use the three months old SD rats, to make higher and more desirable model of Alzheimer’s disease with the Aβ1-42.Use this model to research whether angiotensin converting enzyme inhibitor perindopril can improve the ability of learning and memory of the AD rats.Methods: Three months old SD rats were injected with D- galactose to make the aging model, observe the general condition, changes in body weight, testing SOD, MDA content in blood, compared with the normal aging group of one year old rats. The rats injected with Aβ1-42 in the hippocampus of them to make the AD model, using the Morris water maze test average escape latency of rats in each group and the total distance sailed.Observe the change of the nerve cells in the hippocampus of the rats.Compare the thymus and spleen index of the modle group between three months old and one year old rats. The model group rats were given vascular angiotensin converting enzyme inhibitor and then observe the sailed distance and the effect of perindopril on the expression of iNOS in neuron of hippocampus in AD rats.Results: After subcutaneous injection of D- galactose, SOD levels in the blood in the model group of three months old rats decreased significantly than the control group,MDA level increased significantly.Both P<0.01.Compare to the one year old rats.Three months old of the aging model making success.After the rats injected with Aβ1-42 in the hippocampus of them,using the Morris water maze test average escape latency of rats in each group and the total distance sailed between the three months old and the one year old modle of group,both P<0.01.It have damaged in rat hippocampus of nerve cells,the model of AD indicated success.The results show that the thymus and spleen index of the modle group between three months and one year old rats decreased significantly,P<0.01. After angiotensin converting enzyme inhibitors intervention, the perindopril group rats latency decreased significantly, the number of skip over stage increase, fourth quadrant swimming path of significant grows,iNOS immunoreactive neurons cytoplasm positive expression in the hippocampus decreased obviously, P<0.01,there were statistical significance.Conclusion: The data shows that injecting D- galactose made the aging model use the three months old SD rats and then make them dementia model with the Aβ1-42,are more resistant to the follow experiments for the AD drugs. Angiotensin converting enzyme inhibitor perindopril can improve Alzheimer disease rats learning and memory ability.
