Objectives: To analyze the change of inflammatory mediators IL-1β,TNF-α and growth factor b FGF and TGF-β1 secreted by macrophages on the wound of burned rats under different conditions of hydrogen sulfide,aims to investigate the effects of H2 S on inflammatory response and wound healing after burn.Motheds:78 healthy SD rats were divided into 13 groups(normal control group, normal saline group 1,2,3, Na HS group 1,2,3, PPG group 1,2,3, Na HS+PPG group1,2,3); the normal control group received no intervention, the rest of the groups were formed a model of 5% partial thickness burns, the rats in the normal saline group were timely intraperitoneal injected saline every day, the rats in the Na HS group were timely intraperitoneal injected Na HS solution every day, the rats in PPG group were timely intraperitoneal injected propargylglycine daily, the rats in Na HS+PPG group were timely intraperitoneal injected Na HS solution and propargylglycine daily, on the day of 3,7,14 to take macrophage from the burned rats to culture; applied ELISA to analyze the change of IL-1β, TNF-α, b FGF and TGF-β1concentration in the cell culture supernatant in each group. Analyze the statistical results by one-way ANOVA.Results: At each time point, the concentration of b FGF and TGF-β1were higher than the normal control group, there was statistically significant(P<0.05); at the each time point, the concentration of b FGF and TGF-β1 in Na HS group was higher than that of the corresponding concentration of the saline group, there was statistically significant difference(P<0.05); at the each time point, the concentration of b FGF and TGF-β1 in PPG group was significantly lower than the saline group, there was statistically significant difference(P<0.05); at the each time point, the concentration of b FGF and TGF-β1 in PPG + Na HS group lower than the Na HS group, there was statistically significant difference(P<0.05); the concentration of b FGF and TGF-β1 in PPG + Na HS group was higher than the PPG group at each time point, there was statistically significant differences(P<0.05); the concentration of TNF-α and IL-1β in saline group was higher than normal control group at each time point, there was statistically significant difference(P<0.05); the concentration of TNF-α and IL-1β in Na HS group was lower than the saline group at each time point, there was statistically significant difference(P<0.05); the concentration of TNF-α and IL-1β in PPG group at each time point was higher than the saline group, there was statistically significant differences(P< 0.05); the concentration of TNF-α and IL-1β in PPG + Na HS was higher higher than the corresponding set of concentrations at each time point, there was statistically significant difference(P<0.05); the concentration of TNF-α and IL-1β in PPG +Na HS group was lower than the PPG group corresponds to the concentration at each time point, there was statistically significant difference(P <0.05).Conclusion: Small doses of Na HS could promote wound macrophages secrete b FGF and TGF-β1, at the same time, it inhibit wound macrophages secrete IL-1β and TNF-α, H2 S plays a role of prevention and treatment of wound infection and promote wound healing in the early phase of the burn.
