Tumor Necrosis Factor-αGene Locus 308 And 238 Polymorphisms And Genetic Susceptibilities To Osteoarticular Tuberculosis(OATB) In Tibetan And Han

Objective To investigate the TNF-a promoter polymorphisms in Tibetans people and allele frequencies in this group, we conducted the case-control studies to test the associations between TNF-a-308 and-238 promoter polymorphisms and OATB susceptibility in Tibetan and han.Methods We collected the OATB patients(n=65) and healthy control(n=50) blood and extracted the genome DNA then developed an allele-specific polymerase chain reaction(AS-PCR) assay to identify the TNF-a-238 and-308 locus Polymorphisms, analysis the allele distributions.Results TNF alpha- 308 genotypes and alleles and TNF alpha 238 genotypes and alleles in qinghai Tibetan and han Chinese joint tuberculosis patients and controls the distribution of frequency does not exist significant difference(P > 0.05);Sites of TNF alpha- 308- g/A genotype and allele in qinghai Tibetan, han joint tuberculosis patients and control group comparison difference had statistical significance(P < 0.05);Sites of TNF alpha- 238- g/A genotype and allele in qinghai Tibetan, han joint tuberculosis patients and control group to compare differences had no statistical significance(P > 0.05).Conclusion Not found in qinghai region in Tibetan, the han nationality population TNF alpha- 308- a,- 238- a mutant alleles between chain relationship; And TNF alpha- 308 between Tibetan, the han nationality, no difference- 238 loci genepolymorphism distribution.Not found the TNF alpha- 308-238 loci gene promoter polymorphism and region of qinghai Tibetan, the han people joint tuberculosis associated with genetic susceptibility. Qinghai Tibetan, han TNF alpha- 308 and TNF alpha- 238 loci alleles G and A frequency distribution is not there are racial differences. Joint tuberculosis of genetic susceptibility in qinghai Tibetan and han Chinese there is no genetic diversity.
