Objective:To observe the expression of EGFR and EGFRv Ⅲ in human glioma. Research the relationship between the two and human glioma pathological classification and the correlation between the expressions. Explore their relationship with clinical biological behavior of glioma.established animal model of EGFRVⅢ orthotopic xenografts in nude mice. And ultimately provide theoretical and experimental basic to EGFR and EGFRv Ⅲ targeting therapeutic for GBM in vivoMethods:1 To collect 20 cases of normal brain tissue, 122 cases of glioma specimens;Expressions of EGFR and EGFRv Ⅲ were detected in those cases of glioma and nomal brain tissue by immunohistochemical staining method; Statistical analysis: Experimental data were analyzed using statistical software SPSS 16.0. Use rank sum test(Kraskal-wallis) and Spearman rank correlation analysis. α=0.05 is test criteria.2 Intracranial inoculation using U87- EGFRv Ⅲ cells were transplanted to nude mice.Established orthotopic glioblastoma tumor xenograft animal model of brain tumor-bearing mice were embedded in paraffin using HE staining and immunohistochemical staining of the specimens were stained find tumor microvessel density areas(hot spots) and dyeing clear background well-controlled area, and then photographed at high magnification(10 × 20) and under its biological behavior and heterogeneity analysis.Results:1 Located on the cell membrane of EGFR-positive staining / cytoplasm, brownish yellow granules. The results showed that the expression in normal brain tissue was 15.00%(3/20). In122 cases of human glioma tissue expression was 68.03%(83/122) in normal brain tissues was 15.00%(3/20), differential expression in normal tissues expressing EGFR-grade gliomas with different pathological tissue there was a significant(P <0.01), glioma pathology andexpression level was positively correlated(P <0.01), with higher levels of pathology tended to increase.Statistics glioma tissue samples and other clinical features of 83 cases of cases of EGFR expression in the brain, and survival, KPS score negative correlation(P <0.05), with sex, age, tumor size were not correlated(P > 0.05). EGFRV Ⅲ expression on the cell membrane / cytoplasm / nucleus, brownish yellow granules. EGFRVⅢ in normal brain tissue was negative. EGFRVⅢ nuclear expression in some cases appear.EGFRVⅢ nuclear expression in some cases appear. EGFRVⅢ in 122 cases of human glioma tissue expression was 41.80%(51/122) in normal brain tissue expression was 00.00%(0/20), at different levels of intensity of expression glioma tissue with the increase in pathological level was increasing, and the difference was statistically significant(P <0.05).Statistics glioma tissue samples and other clinical features of 83 cases of cases of EGFR expression in the brain, and survival, KPS score negative correlation(P <0.05), with sex, age,tumor size were not correlated(P > 0.05).
