Schisandrae Chinensis Fructus was the dried fruits of Schisandra chinensis(Turcz.) Baill of Magnoliaceae, also known as“beiwuweizi”. Schisandrae Chinensis Fructus was a well-known tonic and sedative herbal medicine and used as a superior drug in the “Shen Nong Ben Cao Jing”. Schisandrae Chinensis Fructus used as traditional Chinese medicine, has a variety of chemical compositions and pharmacological effects. It could be used with other medicinal materials for treating many diseases. At the same time, Schisandrae Chinensis Fructus which regarded as a food supplements was widely used in food field. Recently, natural resources of Schisandrae Chinensis Fructus have been more and more exhausted because of overexploitation. In addition, The quality control of Schisandrae Chinensis Fructus was still not perfect. Therefore, we carry out the study on chemical compositions of Schisandrae Chinensis Fructus, to compare the difference of lignans content in Schisandrae Chinensis Fructus from different origins, to investigate the distribution of lignans content in different parts(seed kernel, seed shell and pulp) for making the best of its natural resources, to establish the Fingerprints of Schisandrae Chinensis Fructus and different parts. These researches would give some references for evaluating the quality of Schisandrae Chinensis Fructus.The silica gel column and Sephadex LH-20 gel column were applied to isolate the compounds in Schisandrae Chinensis Fructus from the Changbai county of Jilin Province. The five compounds were obtained from petroleum ether parts and was identified by spectroscopic data(1) schisandrin B,(2) β-sitosterol,(3) 1-Octacosanol,(4) gomisin M2,(5) micrantherin A.A stable and reliable HPLC method was developed for quality evaluation of Schisandrae Chinensis Fructus from twelve different origins. Tine lignans, including schisandrol A, gomisin D, gomisin J, schisandrol B, angeloylgomisin H, gomisin G, schisantherin A, schisandrin A, schisandrin B and schisandrin C were determined simultaneously. Ten lignans showed good linearity in linear regression analysis. The recoveries of ten lignans were in the range of 97.46–99.83% with RSD less than 1.98%.In previous studies, the comparison of lignans contents was more concentrated in the leaves, stems, roots, fruit stalk of S. chinensis. There has been relatively little information pertaining to the different parts including seed kernel, seed shell and pulp of Schisandrae Chinensis Fructus. In this study, the lignans contents were determined in seed kernel, seed shell and pulp by the development HPLC mothed. The average content of total lignans of twelve origins for seed kernel, seed shell and pulp were 30.08 mg/g, 9.76 mg/g and 2.24 mg/g, respectively. The total content of the ten lignans in the sample from seed kernel was the highest.The HPLC fingerprints of “Schisandrae Chinensis Fructus and different parts” were established, and the fingerprint of “Schisandrae Chinensis Fructus and different parts” that could clearly express the feature.The in vitro antioxidant activities of pulp, seed shell and seed kernel were investigated using DPPH, ABTS and the total reducing power methods. The scavenging activities decreased in the order of seed shell > seed kernel > pulp. In addition, the antiradical activities of ten lignans(schisandrol A, gomisin D, gomisin J, schisandrol B, angeloylgomisin H, gomisin G, schisantherin A, schisandrin A, schisandrin B and schisandrin C) were also investigated using DPPH and ABTS radical scavenging methods for a preliminary study between structure of lignans and in vitro antioxidant activity. The result indicated that gomisin J which have free phenolic hydroxyl groups shows stronger antiradical activity than other nine lignans which have not phenolic hydroxyl groups.
