Objective The purpose of this study is to investigate the early life of probiotic intervention on rats with allergic rhinitis CD4+CD25+ regulatory T lymphocytes and their impact on the transcription of factor Foxp3+.Method 1.Divide and record the allergic rhinitis group(group A), probiotic intervention group(group B) and the negative control group(group C) of newborn SD rats, pay attention to their scratching nose, foraging, rhinorrhea, sneezing and other acts; 2. To collect the nasal mucosa of rats, give HE staining to them and then observe the Eosinophils infiltration degree; 3.Using ELISA method to examine the serum Ig E levels in each group; 4..Using flow cytometry to analyse the percentage of CD4+CD25+ T cells in the CD4+ T cells; 5.Using Real-time PCR to examine the development of Foxp3 m RNA in rats’ spleen and thymus tissues of the three groups.Results 1. The number of probiotic intervention group rats that have allergic rhinitis was obviously less than the number of the other groups. The difference between them was of statistical significance.(χ2 = 7.2000, P <0.05);2. The score of the Probiotic intervention group symptom score(0.72 ± 0.31) was significantly lower than the allergic rhinitis group(7.61 ± 1.92), the difference was statistically significant(t = 11.2029, P <0.05); 3.The number of the Probiotic intervention serum Ig E(29.91 ± 4.88 ng / ml) group was significantly lower than the allergic rhinitis group(116.21 ± 10.71 ng / ml), and their difference was also of statistical significance(t = 23.1877, P <0.05). But the negative control group(16.81 ± 4.72 ng / ml) showed no significant difference(t = 6.1018, P> 0.05);4. Probiotic intervention group rat spleen and thymus CD4+CD25+ Treg cells in the percentage of CD4+ T cells(12.07±2.87%, 30.81±6.22%) was significantly higher than in allergic rhinitis group(5.46±1.19%, 14.82±3.92%), the difference was statistically significant(t=-6.7278,-6.8775, P<0.05). As to the negative control group(11.05±2.91%, 27.95±5.94%) the difference was not statistically significant(t=0.7892, 1.0516, P>0.05);5. Probiotic intervention group spleen and thymus Foxp3 m RNA expression(20.07 ± 4.89, 18.81 ± 3.64) was significantly higher than in allergic rhinitis group(5.66 ± 2.21,9.81 ± 2.76), the difference was statistically significant(t=-8.49178,-6.2303, P<0.05), with the negative control group(19.75±4.27,17.95±3.51) difference was not statistically significant(t=0.1559,0.5378, P>0.05).Conclusions 1. Probiotic intervention can reduce serum Ig E levels in allergic rhinitis rat models and alleviate type I allergy; 2. Early intervention of probiotics on allergic rhinitis rat model can significantly increase the quantity of CD4+CD25+ regulatory T cells and the expression of Foxp3+ m RNA of e spleen and thymus, then inhibiting the progression of AR rats;...
