Effects Of Rock2 Regulating CEBPD On Proliferation In Hepatocellular Carcinoma

Objective:To investigate the effects of Rho associated coiled coil containing protein kinase2(Rock2) on CCAAT/enhancer-binding protein delta(CEBPD) expresshon affecting tumor proliferation ability in hepatocellular carcinoma(HCC) Methods:1、 The expresshons of Rock2 and CEBPD m RNA were detected by q RT-PCR in 20 cases of HCC tissues, and the correlation between them was analyzed.2、 The expresshons of Rock2 and CEBPD m RNA were detected by Western blot in 20 cases of HCC tissues, and the correlation between them was also analyzed.3、 The m RNA expresshon of Rock2 and CEBPD were detected by q RT-PCR in Rock2 Rock2-sh RNA cells.4、 The protein expresshon of Rock2 and CEBPD were detected by Western blot in Rock2-sh RNA cells.5、 The proliferation abilities of HCCLM3 and SMMC7721 in blank control group, sh Rock2 group and Rock2 restored group were measured by CCK-8 assay and Ed U assay. Results:1、 The results revealed that the average fold change of Rock2 expresshon was shgnificantly increased and that the CEBPD expresshon was shgnificantly decreased in HCC tissues. Moreover, the scatter plots showed that the Rock2 and CEBPD expresshon levels were inversely correlated in HCC tissues.2、 The expresshons of CEBPD m RNA and protein were shgnificantly upregulated in Rock2 knockdown HCC cells.3、 Rock2 regulated HCC cells proliferation abilities through regulating CEBPD expresshon.
