| Objective:To explore the levels of mi R-375 of patients with NSCLC in the serum and to determine whether there was a correlation with clinical parameters,and to analyze the relationship between the levels of mi R-375 and the chemotherapeutic sensitivity of platinum drugs.Methods:1. 82 cases diagnosed with NSCLC at the first affiliated hospital of Nanchang university from September 2013 to September 2014 were included as,100 cases of healthy people were selected from the same period as the control group.2. By RT-PCR measured the level of mi R-375 in exprimental group and control group in serum.3. Explore mi R-375 correlation to clinicopathological parameters of NSCLC patients and its diagnostic efficiency by statistical analysis.4. Advanced(stage III / IV) NSCLC patients in an platinum for drug based chemotherapy after effective(Cr + PR)(n = 16) and ineffective(SD + PD)(n = 22)of mir-375 expression levels were detected by RT-PCR, and the statistical analysis of the sensitivity of chemotherapy.Results:1. The value of 2-ΔΔCтof mi R-375 in experimental group serum was(0.85 +0.19), The value of 2-ΔΔCт of mi R-375 in control group was(1.14 + 0.12). Mi R- 375 in the serum of lung cancer group expression level is lower than normal group(t =12.014,P = 12.014).2. The levels of serum mi R-375 in NSCLC patients’ s TNM Staging and lymphatic metastasis was significantly correlated(P<0.0001,P<0.0001). However, no gender, age, tumor type or diameter, smoking history(P = 0.552, P = 0.255, P = 0.769,P = 0.777, P = 0.212).3. In the diagnosis of NSCLC with ROC area under the curve of serum mi R-375(AUC) 0.905(95%CI: 0.860~0.950),sensitivity and speciality were 92.0% and 78.0% respectively. In the early diagnosis of NSCLC with ROC area under the curve of serum mi R-375(AUC) 0.892(95%CI:0.818~0.967), sensitivity and speciality were98.0% and 29.4% respectively.4. The value 2-ΔΔCтof mir-375 in effective(CR + PR) in serum of patients was(1.06 ± 0.11), The value 2-ΔΔCт of mir-375 in ineffective(SD + PD) in serum of patients was(0.92 ± 0.14), the serum levels of mir-375 with ineffective in patients less than effective in patients(P = 0.0020).Conclusion:1. NSCLC patients were in comparison to control group, the serum levels of mi R-375 was significantly decreased.2. The expression level of mi R-375 was related to lymph node metastasis and TNM staging, and was related to the sensitivity of platinum chemotherapy. |
