Purpose:The aim of this study is to investigate beneficial effect of fenofibrate on retinal degenerationinduced by sodium iodate.Methods:(1)Cell experiment:Immortalized human retinal pigment epithelial cell line(ARPE19 cells) was used for in vitro study.ARPE19 cells were exposed to different concentrations of sodium iodate to determine the does of experiment.ARPE19 cells were exposed to sodium iodate with or without different doses of fenofibrate. Expression of apoptotic protein were determined.(2)Animal experiment:A mouse model of retinal degeneration was set up by intravenous injection of sodium iodate(50mg/kg). 0.1%Fenofibrate were given to mice by daily gavage for three days. To determine the changes of apoptosis in retina and reactive gliosis of Müller cells or inflammatory of retina.Results:(1) Cell experiment:In cultured ARPE19 cells, fenofibrate pretreatment group compared with odium iodate exposure group, the number of TUNEL positive cells has obvious difference(P< 0.01).According to immunofluorescence and western boltting, fenofibrate significantly suppressed sodium iodate-induced expression of cleaved-caspase 3.MTT test showed fenofibrate significantly improve the survival of cells.(2) Animal experiment:Retinal apoptotic cells were increased in mice after sodium iodate injection.The apoptosis of retinal cells has time-dependence and reache peak value on the third day.Administration with fenofibrate significantly inhibited apoptosis of retinal pigment epithelium and photoreceptors in sodium iodate-injected mice(P < 0.01). GFAP expression of Muller cellswas down regulation, inflammatory responserelated proteins CD11 b expression decreased significantly.Conclusions: the results from this study suggested a protective role of fenofibrate on retinal cell survive, which is at least in part mediated by inhibition apoptotic pathway activation.Fenofibrate can reduce reactive gliosis of Müller cells and inflammatory of rentia.
