Purpose: Testing expression of Twist, ZEB1, E Cadherin, Vimentin in normal cervical epithelium, cervical intraepithelial neoplasia(CIN) and cervical cancer and its related to EMT,and analyzing the role of EMT in cervical cancer occurrence, development, invasion and metastasis.Methods: Detected the expression of Twist, ZEB1, E- Cadherin,Vimentin)in 10 samples of normal cervical tissue, 12 cases of CIN I, 18 cases of CINII- III group and 42 cases of cervical cancer tissues by immunohistochemical method(i.e., S- P method)and analysis the correlation between them.Results: 1. The expression rate of E – cadherin was 90% in normal cervical epithelial and the high expression of E – cadherin was 53.41% in CINI, expression of E – cadherin was 44.4% in CINII- III, and the high expression of E – cadherin was 20.00% in the among the cervical squamous carcinoma tissues; While the expression of Vimentin was negative in normal cervical epithelial, and the expression of Vimentin was 8.30% in CINI, the expression of Vimentin was 44.40% in CINII- III, the expressionof Vimentin was 70.0% in cervical cancer tissue and express all have differences between each group, the difference was statistically significant(P < 0.05).2. The high expression of E- cadherin for 4 cases in 5 cases of in cervical cancer tissue of well differentiation, 1 case of lower expression, The expression of E-cadherin was 71.43% in moderate differentiation cervical cancer tissues, the expression of was 52.38% in poor differentiation cervical cancer tissue.Thus,there was significant decrease in E- cadherin in cancers from patients who developed poor differentiation compared with cancers from patients with moderate or well differentiation(P<0.05).In 5 cases of well differentiation, 3 cases was well negative expression of Vimentin and 2 cases was low expression. Expression of Vimentin was 71.40% of moderate differentiation cansers and 80.95% of poor differentiation cervical cancers. The expression of Vim was increased with the decreasing of differentiation level, Thus,there were significant differences between the expression of Vimentin and histologic grade(P<0.05).3. Expression of Twist was 0% in the: normal cervical tissue, Expression of Twist was 41.67% in CINI and 55.56% in CINII- III, Expression of Twist was 60.0% in cervical cancer;There are differences between normal cervix and cervical cancer group and significant on the difference(P<0.05).ZEB1 positive expression mainly located in the cytoplasm, a few in the nucleus,no expression in the normal cervical tissues, expression of ZEB1 was 25.0% in CINI and expression of ZEB1 was 38.89% in CINII-III, expression of ZEB1 was 65.0%. in cervical cancer. Expression of ZEB1 has the difference between different groups and significant the difference(P<0.05).In cervical cancer with lymph node metastasis, ZEB1 and Twist in the cytoplasm nucleus of strong positive rate was higher than cases with lymph node metastasis(P < 0.05)4. Down-regulation of E – cadherin and up-regulation of Vimentin were closely related to histological grade and lymph node metastases of cervical cancer(P<0.05), Expression of ZEB1 related to FIGO stage(P = 0.001) and lymph node metastasis(P<0.05), The expression of Twist associated with cervical lymph node metastases(P < 0.05).5. Statistical analysis of relationship of E cadherin and Vimentin in cervical cancer tissue, both negatively correlated; Increases ZEB1 and Twist may be resulted in the ectopic expression of Vim and rise, and correlated they and Vim were positively And expression of E- Cad showed a negative correlation.Conclusion: Reduced expression of E- cadherin and increased the expression of Vimentin, ZEB1, and Twist in cervical cancer development process played an important role,and were involved in EMT process. EMT, play an important role in the progress of metastasis and progression of cervical squamous cell carcinoma.
