Objective To investigate the inhibitory effects of the dendritic cells loaded with resistant breast cancer antigen on breast cancer in nude mice.Methods Prepared single-cell suspension from the primary breast cancer. Filtered chemotherap-utics using ATP-PCA susceptibility testing system. Treated cancer cells with medium of 1/10IC501/5IC50, 1/2IC50, 1IC50 and 2IC50, until their growth became steady in the medium of 2IC50.Obtained PBMCs from the peripheral blood of the patient with leukapheresis. Adherent cells thus were induced with GM-CSF and IL-4 to generate DCs, which carried cell lysis compounds with resistant strains or cancer cells lysis compounds without drug treatment. The former obtained mature DCs carried resistant breast cancer antigen. Established breast cancer-bearing nude mice model with these resistant strains, randomly divided them into three groups: the treatment group consisted of DCs loaded with resistant breast cancer antigen,the control group consisted of DCs loaded with primary cancer cells antigen, and the blank group consisted of the same volume of normal saline.Observed the changes of the carried cancer and chart the tumor growth curve.Results The lethal effect on cancer cells and the life-prolonging effect on tumour-bearing mice were examined and compared between the experiment group and control group. In Group a(experimental group), T test results between on day 0 and on day 14 statistically significant difference in the tumor volume of mice(v)(t=3.35, p<0.05). The tumor volume on day 0 and day 14 was obviously smaller than the volume on day 0(t=5.21,6.37,P<0.01). In Group b( control group), The tumor volume on day 28 was obviously smaller than day 0(t=4.17,p<0.01). and the difference on day 35 was more obvious(t=5.18,p<0.01). In Group c(blank group) the tumor volume on day 14 and day 28 remarkable increased(t=2.85,3.97) and the difference on day 35 was more obvious(t=4.57,p<0.01). Compared with a and c,b and c,a and b, the tumor volume on day 35 with treatment was significantly different, Group a was the smallest and Group c was the biggest, a<b<c(F=9.65,P<0.01). Compared with a and c, b and c, a and b, weights of tumors on day 35 with treatment weresignificant difference between groups(F=8.95,P<0.01).Conclusion DCs loaded with resistant breast cancer antigen demonstrated significant cancer inhibition effect, in breast cancer-bearing nude mice model, and showed the strongest inhibition effect.
