Objective We investigate the expression of SOCS in peripheral blood mononuclear cells and its correlation with Th17/Treg immune response in children with HSP, to explore the relationship between SOCS and the imbalance of Th17/Treg immunological pathogenesis of HSP.Methods Totally 32 children with Henoch-Sch?nlein purpura were enrolled in the study. Another 20 healthy age-matched children were simultaneously taken as control. Protein levels of CD4+CD+25 、 CD4+IL-17A+ and mean fluorescence intensity(MFI) for phosphorylated-STAT3(p STAT3) protein in CD4+T cells was detected by flow cytometric analysis. The relative expression level of SOCS1 m RNA、SOCS3 m RNA in peripheral blood mononuclear cells was detected by real-time fluorescent PCR. The protein level of IL-6 was assessed by ELISA.Results 1.The levels of IL-6 and MFI p STAT3 protein in CD4+T cells of the HSP children were significantly higher than the control group(t=16.49,P<0.01;t=9.34,P<0.01). 2.The levels of CD4+IL-17A+ in CD4+T cells of the HSP children were significantly higher than the control group(t=7.59,P<0.05), but the levels of CD4+CD25+ in CD4+T cells of the HSP children were lower than the control group(t=4.58,P<0.01). 3. The m RNA expression of SOCS1、SOCS3 in CD4+ cells increased significantly during acute phase of HSP. 4. The m RNA expression of SOCS1、SOCS3 showed a negative correlation with expression of Th17/Treg of the CD4+T cells in HSP children(r=-0.328,P <0.05;r=-0.413,P<0.01).Conclusions There were both IL-6/STAT3 existed in children with HSP and existence of imbalance of Th17/Treg which were the aberrant of Th17 and the insufficient of Treg. There were higher m RNA expression of SOCS1、SOCS3 in CD4+ cells increased significantly during acute phase of HSP and it was a negative correlation with expression of Th17/Treg of the CD4+T cells in HSP. The insufficient of expression level of SOCS1 m RNA、SOCS3 m RNA might lead to abnormal of IL-6/STAT3, It might be associated with the imbalance of Th17/Treg in children with HSP.
