Objective:To investigation the expression of tumor necrosis factor related apoptosis-inducing ligand(TRAIL)and its receptor TRAIL-R2(DR5)and TRAIL-R4 in non-small lung cancer(NSCLC) and its clinical significance.Methods:using enzyme-linked immunosorbent assay detection of the expression levels of 79 cases of NSCLC and 80 cases of normal human TRAIL in the surm;The expression of TRAIL-R2 and TRAIL-R4 detected by immunohistochemistry in 14 cases of NSCLC tissue.Results:1. The expression of TRAIL were lower in maligand NSCLC group than in healthy people(P=0.003<0.05),the expression of TRAIL in NSCLC were closely correlated with clinical stages and pathological grade. the expression of TRAIL in stage III~IV was significantly lower than that in stage I~II(P=0.000<0.05). the expression of TRAIL were positive correlated with differentiation level of NSCLC(P=0.000<0.05). no significant correlated with the pathological types and age and sex.2. The positive expression rate of TRAIL-R2(DR5) and TRAIL-R4(Dc R2) both is93%(13/14) in 14 case NSCLC, were closely correlated with clinical stages and parhological grade. the expression of DR5 in stage III was significantly lower than that instage I~II(P=0.024<0.05).the expression level of TRAIL-R2(DR5) in higher differentian tissue than in the lower(P=0.02<0.05); The expression rate of TRAIL-R4(Dc R2) was also93%(13/14), with the increase in stages, the expression of TRAIL-R4(Dc R2) was higher,the expression of Dc R2 in low differentiation was higher than in high(P=0.034<0.05). 3.There was negative correlation between the expression of TRAIL-R2 and TRAIL-R4 in NSCLC(r=-0.665). no significant correlated with the pathological types and age and sex.Conclusions :1. TRAIL was expression both in normal and NSCLC,the expression level was lower in NSCLC than in normal.2. the expression of TRAIL in NSCLC were closely correlated with clinical stages and pathological grade.3. the expression of TRAIL-R2 and TRAIL-R4 in NSCLC were closely correlated with clinical stages and pathological grade.4. There was negative correlation between the expression of TRAIL-R2 and TRAIL-R4 in NSCLC5. TRAIL and its recepter TRAIL-R2 and TRAIL-R4 may paly important role in genesis and development of NSCLC.6. The expression of TRAIL-R2 provides a target for clinical treatment of NSCLC,TRAIL-R4 play a regulating role in non small cell lung cancer cell apoptosis.
