| Objective: To explore the prognosis affected by the expression of DKK3 and VEGF-C in gastric cancer(GC) and the relation clinical factors of DKK3.Methods: The DKK3 expression and VEGF-C expression in GC tissues and adjacent non-tumor tissues were evaluated by molecular method like PCR, Western Blot. The prognosis affected by DKK3 and VEGF-C was analysis by survival analysisResults: 1. The relative m RNA expression values of DKK3 in adjacent non-tumor tissues were higher than those in GC tissues(DKK3: 0.72±1.16 VS 2.87±1.58,p<0.001).Compared with adjacent non-tumor tissues, VEGF-C in GC tissues were significantly much higher(VEGF-C: 1.35±0.54 VS 0.49±0.20, p<0.001). 2. The protein expression values of DKK3 in GC adjacent non-tumor tissues were lower than those in GCtissues(DKK3: 0.40±0.21 VS 1.27±0.66, p<0.001), VEGF-C in GC adjacent non-tumor tissues were significantly much lower than those in GC tissues VEGF-C: 1.35±0.54 VS 0.49±0.20, p<0.001). 3. 72(40.0%) and 140(77.5%) of 180 GC specimens can be found DKK3 positive expression and VEGF-C positive expression, whereas DKK3 positive expression and VEGF-C positive expression can be 121(67.2%) and 63(35.0%) of 180 surrounding non-tumoral tissue specimens, There were statistically significant differences between GC and adjacent non-tumor tissue specimens.in the positive expression of DKK3(p<0.001) and VEGF-C(p<0.001) 4. In GC tumor tissues there was a significant negative correlation between VEGF-C expression and that of DKK3(p<0.001) 5. Univariate analysis showed location of tumor(p =0.018), tumor size(p =0.018), T staging(p =0.005),N staging(p <0.001),DKK3 expression(p <0.001)and VEGF-C expression(p =0.003)were related to survival. Among them N staging(HR=2.146,p<0.001), tumor size(HR=0.630,p =0.026)and DKK3 expression(HR=0.544,p =0.021)were independent risk factors for survival. 6. There were significantly differences between DKK3 positive expression and DKK3 negative expression in age(p =0.004), N staging tumor(p <0.001),tumor size(p =0.004)and VEGF-C expression(p <0.001)Conclusion: As the independent risk factors, DKK3 expression was related with VEGF-C expression, it also can predict the overall survival of GC, which maybe a promising target genes in gastric cancer. |
