Objective:This study proposed to observe the histological character of whole adult laryngeal tissue serial section by Leica CM3600 XP large freezing microtome, Identify the histologic components of larynx and its boundary, investigate the anatomical delimitation of the larynx and the relationship between ambient tissues, explore the operation method and specification of whole adult laryngeal tissue serial section. This study also investigated the three-dimensional of pathological changes of the laryngeal tissue and characteristics of laryngeal cancer invasion. Leica CM3600 XP large freezing microtome provide more accurate sectional anatomic basis not only of Micro three-dimensional reconstruction of the larynx, but also of CT and MRI diagnosis and range judgment of Laryngeal cancer, The application of Leica CM3600 XP large freezing microtome provides the boundary of safety incisal margin of laryngeal cancer surgery, provides a new thought, method, Technical Exploration and theoretical research of improving the success rate of surgery.Methods:normal laryngeal tissue purchased through formal channels, North China structural int egrity without head and neck disease in normal adult male corpse throat, aged ranges from42 to 56, starting from the hyoid bone, down to the second tracheal rings, two cases with cl early laryngeal anatomic landmarks and normal glottal structurally in fresh frozen. Patholo gy throat tissue was from Shanxi Medical University First Hospital of Otolaryngology Hea d and Neck Surgery in 2013- total 2014 laryngectomy fresh frozen specimens two cases inwith total excision of the larynx, pathology diagnosed as squamous cell carcinoma of the l arynx, all specimens were stored at-80 ℃ freezer.1. Set the parameters of the computer before slicing, the thickness of specimen was 20μm,Slicing speed was 60 mm/s.2. Dispensing of OCT solution. Collection, transportation, preservation, thaw and embedding of specimen.3. Slicing and rubbing.4. HE staining.5. Gathering the picture.Results:1.
