Effect Of Euryale Ferox On The Expression Of SOCS-3、tGF-β1and α-sma In Renal Tissues Of Diabetic Nephropathy Rats

Abstract/Summary:
Objective:To investigate the effect of euryale ferox on the expression of SOCS- 3, TGF-β1 andα-SMA in renal tissues of diabetic nephropathy rats.Methods:55 health clean level male Wister rats were randomly divided into four groups : the normal control group(NC), diabetic nephropathic model group(DN), Euryale ferox treatment group(EF,3.0g/kg/d) and losartan potassium treatment group(LP, 30mg/kg/d).The DN rats models were established by injecting with streptozotocin intraperitoneally.When the DN rats models were established, rats were treated respectively with intragastric administration once daily for 12 weeks.In the same time, the rats in NC group and DN group were given the same volume of distilled water by gavage.Blood serum creatinine,blood urea nitrogen, microalbumin and 24 h urinary protein were me assured after 12 weeks.The renal tissue were processed for HE-stain, Masson-stain and PAS-stain.Immunohistochemisty staining of SOCS-3, TGF-β1 and α-SMA were used to analyze the changes of renal pathology. The m RNA expression of SOCS-3, TGF-β1 and α-SMA were measured by Real-time PCR.Results:(1)Biochemical Indicators compared : At the end of 12 th week, compared with group NC, BUN, Scr, m Alb, 24 h urinary protein were significantly increased in group DN(P<0.05). Compared with group DN, above up-regulations were significantly decreasedin group EF and LP(P<0.05).(2)Pathological changes: compared with group NC, the renal tissue in DN group has glomerular hypertrophy, mesangial area hyperplasia,basement membrane thickening, tubular lumen enlargement, structure out of order(P<0.05). Compared with group DN, in group EF and LP, the pathological changes in renal tissues were significantly alleviated(P<0.05).(3)Immunohistochemistry results : compared with group NC, in group DN, the expression of SOCS-3, in renal tissues were increased significantly(P<0.05); compared with DN group, in group EF and LP, the expression of SOCS-3 was significantly enhanced, meanwhile the expression of TGF-β1 and α-SMA were significantly decreased(P<0.05).(4)Real-time PCR results : compared with the NC group, the expression of SOCS-3 m RNA in renal tissue of DN group were increased, as well as the expression of TGF-β1 m RNA and α-SMA m RNA(P<0.05), compared with group DN, in group EF and LP, the expression of SOCS-3 m RNA was significantly increased, while the expression of TGF-β1 m RNA and α-SMA m RNA were significantly decreased(P<0.05).Conclusions:Euryale ferox may delay the progression of renal interstitial fibrosis in diabetic nephropathy rats and protect renal function by up-regulating the expression of SOCS-3,and down-regulating the expression of TGF-β1 and α-SMA in renal tissues.
