Objective:Through observation at different doses of Euryale ferox salisb drug-containing serum under high glucose intervention of glomerular podocyte nephrin and α-SMA expression, to explore the Possible mechanism protection of Euryale ferox salisb in the kidney.Methods:In vitro culture mice glomerular podocyte as the research objec,according to the human and animal body weight dose conversion factor calculated to calculate the dose,The rats were given euryale ferox salisb decoction to 1.5g/kg·d、3.0g/kg·d and6.0g/kg · d concentration gavage for 7 days.Abdominal aortic blood centrifugal, to obtained the drug-containing serum.After sufficient podocyte culture differentiation, the cells were divided into different groups according to the following groups:(1)Normal group: DEME normal culture medium(sugar contained for 5.5 mmol/L) + normal serum;(2)High glucose group: DMEM high glucose medium(sugar contained for25mmol/L) + normal serum;(3)Benner Pury group: DMEM high glucose medium(sugar contained for 25mmol/L) + Benner Pury;(4)Low-dose euryale ferox salisb group:DMEM high glucose medium(sugar contained for 25mmol/L) +Low-dose euryale ferox salisb drug-containing serum;(5)meduim-dose euryale ferox salisb group:DMEM high glucose medium(sugar contained for 25mmol/L) +meduim-dose euryale ferox salisb drug-containing serum;(6)high-dose euryale ferox salisb group:DMEM high glucosemedium(sugar contained for 25mmol/L) +high-dose euryale ferox salisb drug-containing serum.The podocytes are growned in 37 ℃ for 48 hours.The changes of expression of nephrin and α-SMA were observed by immunofluorescence assay.The gene expression of nephrin and α-SMA m RNA were measured by Real-time PCR.Results:1.The results of immunofluorescence assay :(1)Fluorescence intensity of nephrin in each group:Compared with the normal control group, the high glucose group was obviously reduced, the difference was statistically significant(p<0.05);Compared with the high glucose group, meduim-dose euryale ferox salisb group, high-dose euryale ferox salisb group and Benner Pury group were obviously higher, the difference was statistically significant(p<0.05);Compared with the high glucose group, low-dose euryale ferox salisb group was significantly increased, no significant difference(p>0.05);(2)Fluorescence intensity of α-SMA in each group:Compared with the normal contro group, the high glucose group was significantly higher than that of the normal group, the difference was significant(p<0.05);Compared with the group of high glucose, meduim-dose euryale ferox salisb group, high-dose euryale ferox salisb group and Benner Pury group were significantly reduced, and the difference is significant(p<0.05);Compared with the group of high glucose, low-dose euryale ferox salisb group was not decreased, no significant difference(p>0.05).2.The results of Real-time PCR:(1)The m RNA of nephrin expression in each group:Compared with the normal control group, the high glucose group was obviously reduced, the difference was statistically significant(p<0.05);Compared with the high glucose group, meduim-dose euryale ferox salisb group, high-dose euryale ferox salisb group and Benner Pury group were obviously higher, the difference was statistically significant(p<0.05);Compared with the high glucose group, low-dose euryale ferox salisb group was significantly increased,no significant difference(p>0.05);(2)The m RNA of α-SMA expression in each group:Compared with the normal contro group, the high glucose group was significantlyhigher than that of the normal group, the difference was significant(p<0.05);Compared with the group of high glucose, meduim-dose euryale ferox salisb group, high-dose euryale ferox salisb group and Benner Pury group were significantly reduced, and the difference is significant(p<0.05);Compared with the group of high glucose, low-dose euryale ferox salisb group was not decreased, no significant difference(p>0.05).Conclusion:The Euryale ferox solisb by regulating the expression of nephrin and α-SMA in podocyte, may have some protective effect of kidney.
