Objective:Through examining the roles of b FGF in the directional differentiation of bone marrow mesenchymal stem cells(BMSCs) into leydig cells(Leydigs), this paper explores appropriate approaches by which BMSCs could be directionally differentiated into Leydigs.Methods:The third-generation BMSCs of rats with purification appraisal were inoculated onto a six-orifice board, and then were randomly divided into control group(A), HCG +PDGF induction group(B), HCG + PDGF + 5.0 ng/ml b FGF induction group(C),HCG + PDGF + 10.0 ng/ml b FGF induction group(D) and HCG + PDGF + 20.0 ng/ml b FGF induction group(E). On the 7th, 14 thand 21stday of the research, morphological observation, culture of testosterone level and immunofluorescence test for 3 beta hydroxy steroid dehydrogenase(3 beta HSD)were conducted respectively.Results:1) Cells of B, C, D and E group after induction indicated the features of microscopic were bigger size, being connected with each other in long shuttle or irregular shape,adherent growing next to walls, and having larger and round nucleus. All these characteristics conform to those of Leydigs. 2) With the progress of time and raising concentration of b FGF, testosterone levels for B, C, D and E group gradually went up and positively expressed cells for 3 beta HSD gradually increased. There were statisticalsignificance between C, D, E and B group; as well as between D, E and C group(P <0.05), whereas there were no significance between D and E group(P >0.05).Conclusions:There are clear promoting effects of b FGF in the process of extracorporeal inducted differentiation for BMSC into Levdigs.
