Objective:Observation rat leydig cells after transplantation of changes of serum testosterone levels, explore old SD rat leydig cells transplantation of LOH in rats.Methods:To meet the requirements of LOH rats were divided into two groups: experimental group and control group.The differentiation of Leydig cells in vitro using cell suspension injections Leydig cell transplantation in experimental group rats side of thigh muscles,the number of transplanted cells for 1×106.In 1 d before transplantation, transplantation2, 7, 12, 17, 22, 27 d tail blood method is used to test the serum free testosterone concentrations.Injection of same amount of saline control group the same method, the determination of serum free testosterone concentrations.Results:Through leydig cells transplantation, the experimental group LOH rat serum free testosterone(pg/ml) in 1 d before transplantation, transplantation after 2, 7, 12, 17, 22, 27 d respectively(1.95 + 0.20),(3.51-0.18),(6.31-0.31),(7.22-0.19),(7.47-0.16),(7.49-0.18),(7.54-0.21), with the increase of time, before transplantation, 1 d, 2, 7, 12 d after transplantation testosterone levels increase, the difference was statistically significant, and 17 after transplantation, 22, 27 d difference between serum free testosterone levels have no statistical significance.Control group respectively(1.91 +0.19),(1.95-0.14),(1.92-0.16),(1.93-0.13),(1.95-0.14),(1.91-0.20),(1.96-0.17), therewas no statistically significant difference(P > 0.05).The experimental group compared with control group difference was statistically significant(P < 0.001).Conclusion:Through the leydig cells transplantation, old SD rats serum free testosterone level is significantly increased before transplantation, has good curative effect on old SD rats.
