Objective To investigate the effect of OPN on EMT phenomenon and the biological behavior of Tca-8113 cells. Method 1 Tca-8113 cell lines stably transfected with a plasmid OPN,to establish OPN overexpression Tca-8113 cell lines, and Western Blot and Realtime PCR detect transfect efficiency; 2 Western Blot and cell immunohistochemistry detect OPN, Snail and EMT markers; 3 Cell adhesion assay, migration assay and Transwell invasion assay was used to the observe effect of OPN on cell adhesion, migration and invasion of Tca-8113. Results 1 Tca-8113 cells can stably expressing OPN. 2 Expression of epithelial markers E-cadherin, β-catenin compared with the before transfection OPN is decreased,but mesenchymal markers Vimentin, N-cadherin is increased; Snail expression also increased after transfection. 3 Before transfection, the A value of Tca-8113 cells adhere was(0.41 ± 0.06), the mobility rate of cells is(23.0±5.7)% in 24 h and(49.6±8.6)% in 36h; the number of invasive cells was(16±7.9),after transfection the A value of Tca-8113 cells adhere was(0.61± 0.07), the mobility rate of cells is(44.1±6.3)% in 24 h and(71.6±6.5)% in 36h; the number of invasive cells was(39±6.7), which were increased compared with before transfection,(P<0.05). Conclusion Human OPN is involved in promoting Tca-8113 cells adhesion, migration and invasion by regulating EMT phenomenon, Snail may be promote the function of OPN in this process. |
