| Objective: Intervertebral disc degeneration is an important cause of low back pain, but so far,the physiological and pathological mechanism of IVDD remains unclear. this study intends to testing the different expression quantities of m RNA of inflammatory factor-HMGB-1 In different degradation levels of nucleus pulposus tissue according to Magnetic Resonance Imaging(MRI) Pfirrman classification of the lumbar disc degeneration.Thereby expecting it could be proving that this special inflammation factor has obvious correlation with degree of intervertebral disc degeneration, making this biomarker could be an indicator which can predict the degree of low back pain.It could also helps us to understand the pathogenesis of IVDD, to find suitable targets for the development of targeted therapy drugs, to break new ground in the conservative treatment of LDH, to relieve the pian from patients who suffer Chronic lumbocrural pain.Methods: getting 30 nucleus pulposus specimens from patients’ intervertebral disc who had lumbar posterior open surgery or minimally invasive surgery. The nucleus pulposus specimens were divided into experimental group and control group. The experimental group were further divided into 4 groups according to the Pfirrmann classification method, and the 1,2-5 grade corresponding to the A-D group.The control group were 4 nucleus pulposus specimens from Patients with scoliosis orthopedics. Subsequently observing the expression levels of HMGB-1 by use of Real time-PCR technique, and to measure its expression level of m RNA.The data were analyzed by the one-way ANOVA analysis of SPSS 18.0.Results: The statistical results of HMGB- 1 m RNA expression level according Real time-PCR analysis:the m RNA expression of HMGB-1 were increased progressively with IVDD severity. The one-way ANOVA analysis to the m RNA expression of HMGB-1: compared to the group E between the group B,C and D, the group Abetween the group C and D,the group B and the group C and D,the differences were statistically significance(P<0.05); there was no difference statistically significance among the other groups(P>0.05). AQP3: compared to the group A between the group C, D and E, the group B between the group D and E, the group C and the group E, the differences were statistically significance(P<0.05); there was no the difference statistically significance among the other groups(P>0.05). And statistical results cannot show that the degeneration of nucleus pulposus have significant differences in gender, age.Conclusion:It was significant difference that the gene expression of the HMGB-1 in the normal human nucleus pulposus and the degenerative nucleus pulposus; Compared with the normal nucleus pulposus, the expression of the HMGB-1 was significantly increased in the degenerative nucleus pulposus; and the expression of HMGB-1 in the content was increased progressively with the increasing level of the nucleus pulposus degeneration. |
