Objective:To observe the expression of lysyl oxidase(LOX)in the nucleus pulposus of intervertebral disc with different degenerative grades,and to explore the relationship between LOX and intervertebral disc degeneration.Methods:We collected the nucleus pulposus tissue of human intervertebral disc from March 2018 to December 2018 in our hospital,and the nucleus pulposus tissues of young patients with lumbar fracture who needed anterior decompression and internal fixation were set as the normal control group(group A).The remaining samples were divided into 4 grades according to Pfirrmann classification(corresponding to B,C,D,E groups respectively).After sample treatment,HE staining,imnunohistochemistry,Western blot and PCR test were used to observe the expression of LOX in nucleus pulposus.The SPSS 20.0 statistical software was used to analyze the date.Results:1.HE staining showed that the matrix components were intact,the nucleus pulposus cells were abundant and the cell structure was intact in the normal control group,and the degeneration group proved the contrary:the matrix components were decreased,the nucleus pulposus cells were less and the cell structure was incomplete.2.Immunohistochemistry:the expression rate of LOX positive cells in the degeneration group was significantly lower than that in the control group,and the expression rate of the positive cells was inversely correlated with the Pfirrmann grade.The expression rate of positive cells in each group was analyzed by One-way anova,and the difference was statistically significant(P<0.05).Pearson product moment linear correlation analysis was performed on the expression rate of positive cells of LOX,Pfirrmann grade and age:there was a significant inverse correlation between LOX and Pfirrmann grade(r=-0.9053,P<0.05).Also a significant inverse correlation between LOX and age(r=(-0.9539,P<0.05).3.Western Blot:The expression of LOX protein in the degeneration group was significantly less than that in the control group,and the protein expression of LOX was gradually decreased with the increase of the degeneration degree.One-way anova was performed on the LOX protein expression between the groups,and the difference was statistically significant(P<0.05).4.PCR:The expression of LOX mRNA in the degeneration group was significantly decreased compared with that in the control group,and the expression of LOX mRNA decreased progressively with the increase of the degree of degeneration.One-way anova was performed on the LOX mRNA expression between the groups,and the difference was statistically significant(P<0.05).Conclusion:There is a significant difference in the expression of LOX between the normal and the degenerative nucleus pulposus tissue.The expression of LOX in the degenerative nucleus pulposus tissue was lower than that in the normal nucleus pulposus tissue,and with the aggravation of the degeneration degree of the nucleus pulposus,the expression of LOX decreased gradually.LOX may be involved in the occurrence and development of human intervertebral disc nucleus pulposus degeneration.
