Objective To investigate the expressions of latent membrane protein 1(LMP1)and mucin1(MUC1) in 53 cases of nasopharyngeal carcinomas(38cases with cervical lymph node metastasis, 15 cases without cervical lymph node metastasis)and 15 cases of nasopharyngitis, and evaluate the relationship between each protein and cervical lymph node metastasis, and its role in the development of nasopharyngeal carcinoma and the correlation between the two proteins.Methods In this study, we use the immunohistochemistry to evaluate the expressions of LMP1 and MUC1 in 53 cases of nasopharyngeal carcinomas(38cases with cervical lymph node metastasis, 15 cases without cervical lymph node metastasis) and 15 cases of nasophayngitis, and the Spearman correlation analysis to analyze the relationship of this two proteins, and use SPSS 17.0 statistical analysis software to investigate the roles of those proteins in nasopharyngeal carcinoma.Results LMP1 is mainly expressed in the cytoplasm of nasopharyngeal carcinoma cells, its positive expression is in the area that nasopharyngeal carcinoma cells concentrated. The expression rate of LMP1 in nasopharyngeal carcinoma tissues is 71.70%. There are a small amount of expression in uniform distribution in nasopharyngitis tissues. The expression rate is 20.00%. The difference of expression of LMP1 in nasopharyngeal carcinomas and nasopharyngitises were statistically significantly higher(P<0.01), and the expression of LMP1 in nasopharyngeal carcinomas with cervical lymph node metastasis is apparently higher the in those without cervical lymph node metastasis(P<0.05). Besides, the level of LMP1 in nasopharyngeal carcinoma correlates to the TNM stage(P<0.05), but there is no relationship between the level of LMP1 in nasopharyngeal carcinoma and clinicopathological findings including gender, age, the types of carcinoma.MUC1 is mainly expressed in the nucleus and(or) cytoplasm of nasopharyngeal carcinoma cells, its positive expression as well as LMP1 mainly concentrated in the distribution area of nasopharyngeal carcinoma cells, but in nasopharyngitis it is in uniform distribution. The expression rates of MUC1 in nasopharyngeal carcinoma and nasopharyngitis are 81.13% and 40.00%, the difference of expression is statistically significant(P<0.01). Besides, there is no relationship between the expression of MUC1 in nasopharyngeal carcinoma and its clinicopathological findings including gender, age, the types of carcinoma and the carcinoma with or without cervical lymph node metastasis.Conclusions 1. LMP1 protein existed mainly on cytoplasm and cell membrane of nasopharyngeal carcinoma. The expression level of LMP1 is significantly higher in nasopharyngeal carcinomas than in nasopharyngitises, Besides, the expression of LMP1 correlated to nasopharyngeal carcinoma staging and cervical lymph node metastasis, Which is no relationship with the gender, age, and the type of carcinoma. The above indicate that the LMP1 plays an important role in the occurrence and development of nasopharyngeal carcinoma. 2. MUC1 protein expressed the cell nucleus of nasopharyngeal carcinoma. Besides, MUC1 protein expresses significantly higher in nasopharyngeal carcinomas than in nasopharyngitises. But there is no relationship between the expression in nasopharyngeal carcinoma and its clinicopathological findings including gender, age, the types of carcinoma and the carcinoma with or without cervical lymph node metastasis. 3. The expression of LMP1 and MUC1 in nasopharyngeal carcinoma is positive related.
