| Objective: To observe the expression of CD133, VEGFR1, CXCR4 in the colorectal cancer and its metastatic liver cancer tissues. To confirm the proportion of the CD133+VEGFR1+ cells and CD133+CXCR4+ cells in colorectal cancer tissues. To investigate the possible relationship between CD133+VEGFR1+ cells and CD133+CXCR4+ cells in the hepatic metastasis of colorectal cancer.Methods : 1. The proportion of CD133+VEGFR+ cells and CD133+CXCR4 cells in the colorectal cancer and its hepatic metastasis were analyzed by using FC500 flow cytometry.2.The expression of SDF-1 m RNA and protein in colorectal cancer tissues and hepatic metastasis were investigated by PR-PCR and immunohistochemical technique respectively.Results:1.Flow cytometry showed the proportion of CD133+VEGFR1+ cells was obviously higher in hepatic metastasis than that in primary tumor tissues and the adjacent mormal tissues(P<0.001,P<0.001),there was no statistical differences between primary tumor and the adjacent mormal tissues( P=0.173).2.The proportion of CD133+CXCR4+ cells was significantly higher than in primary tumor and the adjacent mormal tissues(P<0.001;P<0.001), there was no statistical differences between primary tumor and the adjacent mormal tissues similarly( P=0.185).3.There was no regression between the proportion of CD133+VEGFR1+ cells and CD133+CXCR4+ cells in primary tumor tissues and the adjacent mormal tissues( P=0.282,P=0.113).In hepatic metastasis, CD133+CXCR4+ cells increased with CD133+VEGFR1+ cells, while there was no statistical difference(P=0.069).4.The immunohistochemistry showed that positive expression rate of SDF-1 protein in metastatic liver cancer tissues was higher than that of the primary tumor tissue and the adjacent mormal tissues(P<0.001,P=0.001), no statistical differences were showed between primary tumor and the adjacent mormal tissues(P=0.064). 5.The RT-PCR showed that the expression of SDF-1 m RNA was obviously higher than that of the primary tumor tissue and the adjacent mormal tissues(P<0.001,P=0.014),the expression of SDF-1 m RNA was higher in the primary tumor than the adjacent mormal tissues(P=0.005).Conclusion: 1.We evaluated the proportion of CD133+VEGFR1+ cells and CD133+CXCR4+ cells, the expression of SDF-1 were higher in hepatic metastasis than that in primary tumor tissues and the adjacent mormal tissues. 2. The CD133+VEGFR+ hematopoietic progenitor cells plays the leading role in the formation of liver microenvironment of colorectal cancer, it may lead the CD133+CXCR4+ migrating cancer stem cells to the liver to develop hepatic metastasis. |
