Objective:To established successfully rat kidney transplantation models. Then to observe the effect of Notch Ligand Delta-like1 blockade on the survival of rat renal allograft.Methods:The Brown Norway rats as donors, Lewis rats as recipients, to established the allogeneic transplantation models by a combination of the sleeve technique for the artery, a cuff technique for the vein, and the modified technique for the ureter. And according to therapy management, the recipients were divided into two groups: the experimental recipients which were treated with anti-Delta1(HMD1-5) m Abs i.p. at 250 μg at days 0, 2, 4, 6, 8, and 10 after transplantation; the control recipients which gave the same dose of NS at days 0, 2, 4, 6, 8, and 10 after transplantation. Meantime the two groups were divided into 2 subgroups: one subgroup were sacrificed on the 7th day after surgery, and the allograft was resected to make pathological section, and the blood sample taken from IVC to detect the content of cytokine IL-2、IL-4、IL-6、IFN-γ、IL-10. The other subgroup were used to explore the survival time postsurgery, and test the serum creatinine and BUN from the tail vein at day 0, 3, 5 and 7after transplantation.Results:The allograft survival time: compared with control group(7.83±0.31d),the survival time of the experimental group prolonged significantly(9.83±0.47d). The serum creatinine and BUN of the experimental group was lower than that of the controls at concurrent phases. And pathological section: the control group had acute serious rejection, while the other group acute rejection ameliorated. And supernatants from the experiment group contain less level of IL-2、IL-6、IFN-γ, but more IL-4、IL-10.Conclusions:Anti-Delta1 therapy could prolong the survival time of renal allograft significantly,result from the Th1-Th2 axis converse into the Th2.
