The Research On Analyzing Differentially Expression Genes In Hepatic Echinoceccosis Of Liver Tissue By Using Microarray

Echinococcosis is a common people and comorbidities,which mainly distributed in animal husbandry developed central and western regions of China.Seriously affect the local economy and social development. Interactions between host- the parasite is very complex,the results may be parasite continues to grow and develop or necrosis,or may cause the fibrosis of the liver cells, apoptosis, or regeneration. Hepatic hydatidosis Clinical hidden period for about 5-10 years,Early diagnosis is difficult, clinical course and complex, diverse clinical characteristics. At present, the basis studies of echinococcosis in animal experiments or certain genetic researches.Objective: Using Microarray technology to screen differentially expression genes in liver tissues between different clinical characteristics of Hepatic Hydatid, and lay the foundationt to explore the pathogenesis of Echinococcosis and related basic research.Methods:Select 10 cases of hepatic hydatid surgery cases from June 2013 to December 2014 in Ning Xia Region,collect normal liver tissue after close to intraoperative trim ectocyst outside the capsule wall.Adopts the Gene Chip® Prime View? contains 3.5 million genes of human gene expression profile chip. Divided the genes and the 10 cases of sample into A, B, C, D four groups according to different clinical features, and to find groups of differentially expressed genes by using SAM(Significance analysis of microarray) software;and carried on the preliminary analysis on its Biological processes involved in according to the Gene Ontology(GO) of Biological genetic screening Process(in Process) classification.Results : This experiment screened 78 differentially expressed genes betweendifferent clinical characteristics,up-regulation 72, down-regulation 6,involving complement activation and immunity, inflammation, fibrosis, and calcification, signal transduction and regulation,such as material synthesis metabolism biological processes.Conclusion: 1.Gene chip technology is a new kind of molecular biology techniques, the low consumption, high sensitivity, high throughput analysis of the characteristics of gene expression spectrum in the cell, and its extensive application prospect has caused widespread concern in our country; 2.And liver echinococcosis differentially expressed genes between different clinical characteristics involved complement activation and immunity, inflammation and fibrosis, calcification, signal transduction and regulation, such as material anabolic biological processes. 3.Patients with hepatic hydatid disease of liver tissue fibrosis, and calcification, inflammation, hydatid inner bladder infections, sac fistula formation is closely related to the development and changes between its clinical course. 4.Many factors of the host participate in the reaction,these factors involve in the molecular pathways may be:Wnt signaling pathway( ko04310); MAPK signaling pathway(ko04010);P53 signaling pathway(ko04115);Intestinal immune network for Ig A production(ko04672);Complement and coagulation cascades(ko04610). 5.This topic screening of differentially expressed genes, and the correlation between and signaling pathways involved, and will lay a foundation for further study the pathogenesis of hepatic echinococcosis. 6.On the basis of this topic, can further between normal people and patients with hepatic echinococcosis differences of gene expression profile of research.
