| Objective To study the effects of Acupuncturing Back-Shu and Front-Mu Points on acute renal failure induced by glycerol in rats.Methods 1. Establishing an animal model of acute renal failure in ratsNinety healthy male specific pathogen free(SPF) SD rats were randomly divided into nine groups(10 rats /each group): 0h,1h,3h,6h,12 h,1d,2d,3d and 5d. A week after adaptive feeding, rats were dehydrated for 24 h before glycerol injection(8 ml/kg) in their hind limbs. Blood and kidney samples were collected at 0h,1h,3h,6h,12 h,1d,2d,3d,5d. The levels of serum creatinine, urea nitrogen and Cystatin C were determined and pathological changes in kidney were examined by light microscopy. 2. Effects of acupuncturing large intestine Back-Shu and Front-Mu Points against acute kidney injury in ratsFifty healthy male specific pathogen free(SPF) SD rats were randomly divided into 4 groups(10 rats /each group): model group, colon dialysis group, acupuncturing Back-Shu and Front-Mu Points group, combined therapy group. Rats were dehydrated for 24 h before glycerol injection. Colon dialysis group were given intramuscular injections of 50% glycerol(8 ml/kg) in their hind limbs after 2d pretreatment with colon dialysis. Acupuncturing Back-Shu and Front-Mu Points group was given glycerol after 2 d pretreatment with acupuncturing Back-Shu and Front-Mu Points. Combined therapy group was given glycerol after 2 d pretreatment with the two treatments. The rats were sacrificed three days after intramuscular injection. The levels of serum creatinine, urea nitrogen and Cystatin C were determined and pathological changes in kidney were examined.Results 1. Establishing an animal model of acute renal failure in ratsâ‘ After intramuscular glycerol injection into male SD rats, serum creatinine and urea nitrogen maximally increased at 3 to 72 h and then decreased at 5 day. â‘¡CK was significantly increased at 1 to 3h and then decreased at 24 h. â‘¢The basic histologic abnormalities were renal tubular epithelial cell degeneration,tubular necrosis, cast formation, brush border loss. 2. Effects of acupuncturing large intestine Back-Shu and Front-Mu Points against acute kidney injury in ratsâ‘ The serum creatinineã€urea nitrogen and Cystatin C level in three treatment group were significantly lower than that in model group. There was no significant difference between the 3 treatment groups. â‘¡In the model group, the basic histologic abnormalities were tubular necrosis, cast formation. Compared with the model group, tubular cell necrosis and cast formation partially decreased in 3 treatment groups.â‘¢ Kidney coefficients in colon dialysis group were significantly lower than that in model group. Kidney coefficients in combined therapy group and acupuncturing Back-Shu and Front-Mu Points group decreased a little compared with the model group, and there was no statistically significant in the differences.Conclusion The results suggest that acupuncturing large intestine Back-Shu and Front-Mu Points have protective effects against glycerol-induced acute kidney injury in rats by decreasing the level of BUN, SCr, and Cys C and reducing the degree of the renal tubular damage. Acupuncture treatment have a good effect on removing uremic toxins through gastrointestinal way and it provides a new treatment option for renal failure patients. |
